Buildup from birth onward of short telomeres in human hematopoietic cells

Summary: Researchers used the TeSLA method to measure the accumulation of short telomeres in human hematopoietic cells and found that this accumulation is age-dependent. The proportion of short telomeres is higher in hematopoietic cells of patients with dyskeratosis congenita-telomere biology disorders (DC/TBDs) compared to the general population. Overall, hematopoietic cell telomeres are unlikely to reach the replicative limit during the lifespan of most contemporary humans.

The distribution and accumulation of the shortest telomeres in telomere biology disorders

Summary: Individuals with telomere biology disorders (TBDs) have significantly shortened telomeres in blood cells, which is associated with increased risk of bone marrow failure and reduced survival. The mean telomere length is estimated to be 5 kilobases, but it is the shortest telomeres that indicate cellular senescence. By using the Telomere Shortest Length Assay (TeSLA), it was found that TBD patients have much shorter mean telomere length and an increased number of telomeres less than 3 kilobases. In addition, TBD patients with severe bone marrow failure and multiple organ manifestations have even shorter telomeres.

Summary: This study investigated the relationship between CHIP, LTL, and CAD using data from the Trans-Omics for Precision Medicine (TOPMed) program and UK Biobank. The results showed that genetically imputed longer LTL increased the risk of developing CHIP, which then hastened the shortening of measured LTL. There was also evidence of mediation between CHIP and CAD through LTL.
