Interrupted Nef and Meyer Reactions: A Growing Point for Diversity-Oriented Synthesis Based on Nitro Compounds

Summary: This review summarizes the Nef and Meyer reactions as key transformations of aliphatic nitro compounds, with a focus on the interrupted versions in which the normal pathway is redirected by an external nucleophile. These processes greatly enhance the synthetic potential of nitro compounds. The prospects of this methodology in diversity-oriented organic synthesis are analyzed. The bibliography includes 90 references.

Comprehensive Assessment of Enantioselective Bioactivity, Toxicity, and Dissipation in Soil of the Chiral Herbicide Flurtamone

Summary: In this study, the enantioselective biological effects of flurtamone enantiomers were investigated. The bioactivities of R-flurtamone were significantly higher than S- and Rac-flurtamone, with the R-enantiomer contributing the most to herbicidal activity. Molecular docking revealed the binding energies and interactions of flurtamone and its enantiomers with phytoene desaturase (PDS). Furthermore, the toxicity levels to Selenastrum capricornutum showed that R- and Rac-flurtamone were highly toxic, while S-flurtamone had moderate toxicity. Overall, developing R-flurtamone can reduce environmental risks and protect human health.

Population Pharmacokinetic and Pharmacodynamic Analysis of Polmacoxib in Healthy Volunteers and Patients With Osteoarthritis

Summary: This study explores the population pharmacokinetic and pharmacodynamic characteristics of polmacoxib, a new coxib. The pharmacokinetic properties were described by a 2-compartment model with mixed absorption kinetics. The clearance of polmacoxib was influenced by iron concentration. The relationship between polmacoxib concentration and WOMAC was best described by a 2-effect compartment model, with a required concentration of 508 ng/mL for 50% of the maximum effect.

Interrupted Nef Reaction of Cyclic Nitronates: Diastereoselective Access to Densely Substituted α-Chloronitroso Compounds

The research also highlights the utility and potential of allenic esters as C2 synthons and 1,2-biselectrophiles in cycloaddition reactions.

Controllable Access to Furans and Dihydrofurans through Cyclization/Coupling of Internal Acetylenic β-Ketoesters with Aryl Bromides

Asymmetric Synthesis of a Pyrrolizidinone-Based hNK1 Antagonist through Reductive Ring Contraction of a Six-Membered Cyclic Nitronate

Summary: A concise seven-step asymmetric synthesis method was developed for the synthesis of MSD's potent hNK(1) antagonist, which contains a pyrrolizidinone core bearing two fluorine-substituted aryl groups. The pyrrolizidinone unit was constructed by reductive recyclization of a properly functionalized cyclic nitronate, which was assembled by stereoselective [4+2]-cycloaddition. X-ray analysis confirmed the previously suggested configuration of the hNK(1) antagonist based on bioactivity data, and revealed multiple weak interactions involving fluorine atoms in the crystal structure.

Chemodivergent Photocatalytic Synthesis of Dihydrofurans and β,γ-Unsaturated Ketones

Summary: A new synthetic method has been developed for the generation of 2,3-dihydrofurans or beta,gamma-unsaturated ketones starting from alpha-halo ketones and alkenes catalysed by Ir(ppy)(3) under visible-light irradiation. The redox-neutral nature of the process and mild reaction conditions make it sustainable by avoiding the use of sacrificial reactants and stoichiometric strong oxidants. Detailed mechanistic pathways have been disclosed through experimental investigations and DFT calculations, showing that the synthesis can be directed towards different products depending on the substrates and experimental conditions chosen.
