Chiral Auxiliary-Directed Site-Selective Deprotonation of the Cubane Skeleton

Summary: The first diastereoselective synthesis of trisubstituted cubanes was achieved using a chiral auxiliary. At least three substituents must be introduced at the appropriate positions to establish chirality within the cubane skeleton. The ready conversion of cubane carboxylic acid to a chiral amide, followed by sequential ortho-selective deprotonations and electrophilic trapping, resulted in the formation of 1,2,3-trisubstituted cubanes with high diastereoselectivity. This new route offers potential for the preparation of enantio-enriched cubanes.

Rapid and Scalable Halosulfonylation of Strain-Release Reagents

General access to cubanes as benzene bioisosteres

Summary: The replacement of benzene rings with sp(3)-hybridized bioisosteres in drug candidates generally improves pharmacokinetic properties while retaining biological activity. Rigid, strained frameworks such as bicyclo[1.1.1]pentane and cubane are particularly well suited as the ring strain imparts high bond strength and thus metabolic stability on their C-H bonds. We report expedient routes to 1,3- and 1,2-disubstituted cubane building blocks using a convenient cyclobutadiene precursor and a photolytic C-H carboxylation reaction, respectively, and develop cross-coupling protocols using copper as a catalyst. Our research enables facile elaboration of all cubane isomers into drug candidates, thus enabling ideal bioisosteric replacement of ortho-, meta-, and para-substituted benzenes.

A catalytic alkene insertion approach to bicyclo[2.1.1]hexane bioisosteres

Summary: A general synthetic route to substituted bicyclo[2.1.1]hexanes (BCHs) has been developed through intermolecular coupling between olefins and bicyclo[1.1.0]butyl (BCB) ketones. The SmI2-catalysed process can be applied to a wide range of electron-deficient alkenes and substituted BCB ketones, with low loadings of SmI2. The resulting BCH ketones have been shown to be versatile synthetic intermediates and can be used for selective downstream manipulation and the expedient synthesis of a saturated hydrocarbon analogue of the broad-spectrum antimicrobial, phthalylsulfathiazole.

Strain-Release [2π+2σ] Cycloadditions for the Synthesis of Bicyclo[2.1.1]hexanes Initiated by Energy Transfer

Summary: Saturated bicycles are increasingly important in the design and development of new pharmaceuticals. This article describes a new strategy for synthesizing bicyclo[2.1.1]hexanes, which have defined exit vectors but are underexplored as building blocks in terms of substitution patterns. The process involves sensitization of a bicyclo[1.1.0]butane followed by cycloaddition with an alkene. The scope and mechanistic details of the method are discussed.

Rings in Clinical Trials and Drugs: Present and Future

Summary: In this study, we conducted a comprehensive analysis of ring compounds and FDA-approved drugs. We found that 67% of small molecules in clinical trials only utilize ring systems that are already present in marketed drugs. Additionally, there are approximately 450,000 unique ring systems derived from synthesized chemical space, but molecules in clinical trials represent only 0.1% of this pool. Furthermore, by making systematic changes to existing drug and clinical trial ring systems, we were able to predict potential future clinical trial ring systems that could cover a significant portion of novel ring systems entering clinical trials.

Synthesis of meta-substituted arene bioisosteres from [3.1.1]propellane

Summary: Small-ring cage hydrocarbons are popular bioisosteres for para-substituted benzene rings in drug design. In this study, we synthesized hydrocarbons called bicyclo[3.1.1]heptanes (BCHeps) from [3.1.1]propellane, which have geometric correspondence to meta-substituted benzenes. The BCHeps can be easily transformed into medicinally relevant carbon- and heteroatom-substituted compounds, displaying enhanced metabolic stability compared to their parent arene-containing drugs. This research demonstrates the potential of BCHeps as sp(3)-rich motifs in drug design and provides a new surrogate for meta-substituted benzene rings in drug discovery programs.

Small rings in the bigger picture: ring expansion of three- and four-membered rings to access larger all-carbon cyclic systems
