Summary: Chemiluminescence with minimal interference has been applied in biosensing and bioimaging. However, the emission of traditional chemiluminescence probes limits resolution and penetration in biological objects. This study developed a H2S-activated NIR-II chemiluminescence probe with long-duration imaging, deeper tissue penetration, and specific H2S response. The probe also showed detection capability for metformin-induced hepatotoxicity with higher signal-to-background ratios than NIR-II fluorescence mode.

Summary: Gas-based therapy using virus-mimicking hollow mesoporous tetrasulfide-doped organosilica as a nano-adjuvant shows potential for enhancing anti-tumor immune response in preclinical breast cancer models. The gas nanoadjuvant achieves tumor-specific drug release, promotes photodynamic therapy, and produces H2S and CO as gas immunoadjuvants to activate the cGAS-STING pathway. This approach potentiates photoimmunotherapy of poorly immunogenic breast tumors.

Summary: Despite the initial promise of photothermal ablation (PTA) therapy, the inability to completely ablate large tumors is problematic. This study developed a smart photothermal nanosystem (PBM) based on FDA-approved Prussian blue (PB) nanoparticles, doped with Mn (III) to address this issue. The PBM nanosystem amplified the PTA-induced cGAS-STING pathway, leading to a robust immune response and suppression of both local residual and distant tumors.

Summary: Cancer research has made significant progress in the past few decades, and one promising development is the use of aggregation-induced emission luminogens (AIEgens) as molecular imaging and therapeutic agents. AIEgens have unique properties such as large Stokes shift, high quantum yield, biocompatibility, and photostability, making them suitable for various cancer targeting strategies. They have shown great potential in fluorescence imaging, especially near-infrared imaging, for reliable in vivo cancer imaging.

Summary: The development of rapid, selective, and sensitive fluorescent sensors is crucial for visualizing and quantifying biological molecules and processes in various scenarios, which has significant implications in both basic biological studies and accurate disease diagnosis. Activity-based sensing (ABS) using molecular reactivity as a method for analyte detection offers advantages such as high specificity, sensitivity, and accuracy. However, most conventional fluorophores face limitations due to the aggregation caused quenching phenomenon, which leads to reduced labeling efficiency and low photobleaching resistance. In contrast, aggregation induced emission (AIE) active luminogens (AIEgens) provide exceptional molecular frameworks for ABS, with click chemistry being a popular choice. This review discusses the principles of ABS with AIEgens and click chemistry, and provides a comprehensive summary of the application of specific click chemistry reactions in AIEgen ABS protocols for detecting various target analytes, as well as showcasing the utility of click chemistry in constructing AIEgens for bioimaging.

Summary: In this study, M@P@HA nanoparticles were used as a STING signal amplifier to enhance innate immunotherapy in tumors. The nanoparticles released reactive oxygen species under light irradiation, disrupting cellular redox homeostasis and initiating the STING signal through damaged mitochondrial dsDNA. The immunoregulator Mn2+ increased the activity of cGAS and STING, further amplifying the STING signal in tumor cells. Additionally, the STING signal of TAMs was activated by capturing dsDNA and Mn2+ that escaped from tumor cells, enhancing innate immunity.
