Calcium homeostasis and cancer: insights from endoplasmic reticulum-centered organelle communications

Shanliang Zheng et al.

Summary: Calcium ion (Ca2+) is a widely present and versatile signaling molecule that controls various cellular processes essential for cancer establishment. The dysregulation of Ca2+ homeostasis, particularly in the endoplasmic reticulum (ER), has been considered as a major driving force in the initiation and progression of malignant diseases. This article discusses the ER-centered dysregulation of Ca2+ homeostasis in cancer, summarizes Ca2+-based anticancer therapeutics, and emphasizes the significance of further understanding the regulation of Ca2+ homeostasis in cancer.

PINK1/Parkin-mediated mitophagy inhibits osteoblast apoptosis induced by advanced oxidation protein products

Wei Li et al.

Summary: Osteoblast apoptosis is important in age-related bone loss and osteoporosis, and our study showed that AOPPs can induce ROS production, depolarize mitochondrial membrane potential, trigger apoptosis, and lead to osteoblast apoptosis and bone damage. Furthermore, AOPPs also induce mtROS generation, aggravating oxidative stress and promoting apoptosis. However, treating AOPP-induced apoptosis by activating mitophagy and preserving mitochondrial function may be a potential therapeutic strategy for age-related osteoporosis.

Non-muscle myosin II isoforms orchestrate substrate stiffness sensing to promote cancer cell contractility and migration

Yueting Peng et al.

Summary: The study found that NMIIA plays a role in establishing cell polarity, while NMIIB is involved in generating traction force and polarized distribution, depending on substrate stiffness. Substrate stiffness modulates the distribution and activation of NMII isoforms through multiple signaling pathways and kinase-specific phosphoregulation.

GRP75-faciliated Mitochondria-associated ER Membrane (MAM) Integrity controls Cisplatin-resistance in Ovarian Cancer Patients

Jing Li et al.

Summary: GRP75-faciliated MAM formation is enriched in cisplatin-resistant ovarian cancer cells. Diminishing MAM integrity by GRP75-deficiency reduces ER-to-mitochondrial Ca2+ transfer, accelerates mitochondrial dysfunction and ROS accumulation, and enhances cisplatin-triggered apoptotic cell death.

Drp1-dependent mitochondrial fission in cardiovascular disease

Jia-yu Jin et al.

Summary: Drp1 is a key pro-fission protein involved in the regulation of mitochondrial dynamics through various posttranslational modifications, playing a crucial role in the dynamics of heart cells and organs. Maintaining mitochondrial homeostasis is essential for the contractile function and metabolism of the heart, while defects in mitochondrial dynamics are a critical part of the pathophysiology underlying various cardiovascular diseases.

Tumour-associated macrophages drive stromal cell-dependent collagen crosslinking and stiffening to promote breast cancer aggression

Ori Maller et al.

Summary: Stromal stiffening in malignant tumours is associated with poor treatment outcomes and increased tumour aggression. Identifying factors that regulate stromal stiffening could lead to biomarkers for patient stratification and interventions to improve outcomes. Experimental studies have shown that therapeutic targeting of tumour-associated macrophages and collagen crosslinking enzymes can reduce metastasis and stromal stiffening in aggressive breast cancer subtypes with stiff stroma. Additionally, the expression of lysyl hydroxylase 2 in the stroma has been linked to disease-specific mortality in breast cancer patients, indicating its potential as a stromal biomarker.

Autophagy in host stromal fibroblasts supports tumor desmoplasia

Jenny A. Rudnick et al.

Summary: Growing evidence suggests that autophagy in the host stroma plays a crucial role in influencing the tumor microenvironment, particularly in promoting mammary tumor progression. Loss of fibroblast autophagy hinders the secretion of type 1 collagen, leading to an impairment in the development of a stiff tissue matrix conducive to mammary tumor growth. Therefore, targeting autophagy in stromal fibroblasts could be a potential strategy to inhibit the desmoplastic response required for cancer progression.
