Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Venetoclax imparts distinct cell death sensitivity and adaptivity patterns in T cells

Lindsey M. Ludwig et al.

Summary: BH3 mimetics, particularly the BCL-2 specific inhibitor venetoclax (ABT-199), have shown potential as anti-cancer therapeutics and also impact the immune system, particularly T cell subsets. Short-term BCL-2 inhibition leads to changes in T cell populations towards a more memory cell state, while long-term blockade does not significantly alter T cell landscape.

Polymersomes Decorated with the SARS-CoV-2 Spike Protein Receptor-Binding Domain Elicit Robust Humoral and Cellular Immunity

Lisa R. Volpatti et al.

Summary: The study found that decorating multiple copies of the SARS-CoV-2 spike protein receptor-binding domain (RBD) on the surface of polymersomes can promote a potent neutralizing antibody response. Both RBDsurf and RBDencap induce robust T cell immune responses, producing multiple Th1-type cytokines.
