Quick and Mild Isolation of Intact Lysosomes Using Magnetic-Plasmonic Hybrid Nanoparticles

Summary: The efficient isolation of intact lysosomes is crucial for studying their functions and metabolites. A nanoparticle-based magnetic separation method was developed to target lysosomes using magnetic-plasmonic hybrid nanoparticles conjugated with amino dextran. The intracellular transport process of these nanoparticles was analyzed to determine the optimal conditions for magnetic isolation. The study found that the intactness of lysosomes can be quickly impaired, and optimal settings for high purity isolation were identified.

Targeting Magnetic Nanoparticles in Physiologically Mimicking Tissue Microenvironment

Summary: In this study, we fabricate a biomimetic microvasculature in a flexible tissue phantom and demonstrate distinctive mechanisms of magnetic-field-assisted controllable penetration of biocompatible iron oxide nanoparticles across the same. Our findings suggest that the flexibility of microvascular pathways, magnetic pull, and viscous friction play a crucial role in optimizing vascular penetration and targeting efficacy. These findings have important implications for personalized targeted therapies based on individual vascular properties.

Closed-Loop Temperature-Controlled Magnetic Hyperthermia Therapy with Magnetic Guidance of Superparamagnetic Iron-Oxide Nanoparticles

Summary: This study proposes a closed-loop temperature controller for targeted hyperthermia therapy using superparamagnetic iron oxide nanoparticles (SPIONs). Accurate temperature control is achieved by adjusting the magnetic field strength. The SPIONs can be actively manipulated towards the target using a rotating magnetic field (RMF), followed by exposure to an alternating magnetic field (AMF) for controlled heating. In vitro and in vivo experiments demonstrate the effectiveness of SPIONs in reducing cancer cell viability and tumor volume.
