Platelet-derived extracellular vesicles ameliorate intervertebral disc degeneration by alleviating mitochondrial dysfunction

Zhanqiu Dai et al.

Summary: Mitochondrial dysfunction is a key factor causing intervertebral disc degeneration (IVDD), which involves the production of reactive oxygen species (ROS) and oxidative damage. Platelet-derived extra-cellular vesicles (PEVs) have anti-inflammatory effects and can restore impaired mitochondrial function. In this study, we investigated the potential therapeutic effects of PEVs on IVDD in a rat model and found that they can reduce oxidative stress and slow down the progression of IVDD.

Aligne d lovastatin-loade d electrospun nanofib ers regulate collagen organization and reduce scar formation

Zuhan Chen et al.

Summary: In this study, we fabricated electrospun nanofiber membranes of polycaprolactone/silk fibroin with or without lovastatin loading, and evaluated their scar-inhibitory effects on wounds. The nanofiber membranes demonstrated good controlled-release performance, mechanical properties, hydrophilicity, and biocompatibility. The perpendicular placement of nanofibers to the tension direction effectively reduced scar formation and promoted skin regeneration, while lovastatin-loaded nanofibers inhibited myofibroblast differentiation and migration, synergistically reducing scar formation.

Bioprinted constructs that simulate nerve-bone crosstalk to improve microenvironment for bone repair

Tianchang Wang et al.

Summary: The cross-talk between nerves and bone is crucial for bone repair. Schwann cells play a vital role in regulating the microenvironment through exosomes. This study confirms that Schwann cell-derived exosomes promote bone regeneration by regulating the TGF-beta signaling pathway and improving the microenvironment by promoting innervation, vascularization, and osteogenesis. Engineered constructs that simulate the nerve-bone crosstalk were developed, providing a new strategy for bone repair.

Decline in IGF1 in the bone marrow microenvironment initiates hematopoietic stem cell aging

Kira Young et al.

Summary: As individuals age, the function of hematopoietic stem cells (HSCs) declines, leading to limited health span of the blood and immune systems. Research indicates that the hallmarks of aging in HSCs and hematopoiesis start accumulating by middle age, with decreased levels of the longevity-associated molecule IGF1 in the bone marrow microenvironment playing a key role in causing HSC aging. By directly stimulating middle-aged HSCs with IGF1, molecular and functional hallmarks of aging can be rescued, highlighting the complex relationship between IGF1 levels, longevity, and HSC function.
