Self-Assembly of Glycerol-Amphiphilic Janus Dendrimers Amplifies and Indicates Principles for the Selection of Stereochemistry by Biological Membranes

Dapeng Zhang et al.

Summary: The principles for the selection of the stereo-chemistry of phospholipids of biological membranes are still unclear, and new experiments in this field can help make progress. Three libraries of glycerol-amphiphilic Janus dendrimers (JDs) were synthesized to study the self-assembly of these molecules and the formation of dendrimersomes (DSs). The results showed that non-symmetric homochiral JDs mainly formed unilamellar DSs, while racemic JDs favored multilamellar DSs, indicating a new rationale for chiral selection and suggesting the preference for symmetric achiral lipid assemblies in prebiotic cell membranes.

Carbon dots nanophotosensitizers with tunable reactive oxygen species generation for mitochondrion-targeted type I/II photodynamic therapy

Yunxiu Zhang et al.

Summary: In this study, three types of red-emissive carbon dots (RCDs) were successfully prepared with maximum emission at approximately 680 nm. The RCDs exhibited adjustable generation of reactive oxygen species (ROS) through type I and type II photodynamic therapy (PDT). The unique ROS formation of RCDs was determined by their core sizes and surface states, and they were able to induce cell programmed death via activating mitochondrion-mediated apoptotic pathways. This work provides a new prospect for RCDs as nanophotosensitizers to overcome the limitations of single type PDT.

A Supramolecular Strategy to Engineering a Non-photobleaching and Near-Infrared Absorbing Nano-J-Aggregate for Efficient Photothermal Therapy

Meihui Su et al.

Summary: A supramolecular strategy was presented to build a water-stable, nonphotobleaching, and NIR-absorbing nano-PTA (J-NP) from J-aggregation of halogenated BODIPY dyes for efficient in vivo photothermal therapy. The remodeled J-NP significantly improved cellular uptake efficiency and exhibited efficient antitumor photothermal activities in vivo. This work not only establishes a supramolecular platform for high photothermal performance PTAs but also enables appealing optical functions derived from J-aggregation to be applied in vivo.

Molecular Mechanisms Related to Oxidative Stress in Retinitis Pigmentosa

Carla Enrica Gallenga et al.

Summary: Retinitis pigmentosa is a genetic retinopathy, but non-genetic factors such as inflammation, autophagy, and oxidative stress play a crucial role in its pathogenesis and progression. Oxidative stress, caused by constant exposure to light/oxidative stress, results in accumulation of ROS in the retina, leading to microglial activation and photoreceptor degeneration. Various genetic mutations and alterations in cellular response contribute to the vicious cycle of neuroinflammation and degeneration in RP, suggesting microglia as a key target for oxidative stress therapy.

Overcoming barriers in photodynamic therapy harnessing nano-formulation strategies

Jianlei Xie et al.

Summary: Photodynamic therapy (PDT) has shown great potential in tumor treatment due to its non-invasiveness, spatiotemporal selectivity, lower side-effects, and immune activation ability. However, the clinical application of PDT is restricted by various factors such as traditional photosensitizer limitations, light penetration issues, tumor resistance, and pain induction. Recent advancements in using nanomaterials as photosensitizers or carriers, combined with optimized therapy strategies, have shown promising results in overcoming these barriers and improving the effectiveness of PDT in cancer treatment.
