ATP-Coated Dual-Functionalized Titanium(IV) IMAC Material for Simultaneous Enrichment and Separation of Glycopeptides and Phosphopeptides

Danqing Wang et al.

Summary: A hydrophilicity-enhanced bifunctional Ti-IMAC material with grafted adenosine triphosphate (denoted as epoxy-ATP-Ti4+) was developed for simultaneous enrichment and separation of common N-glycopeptides, phosphopeptides, and M6P glycopeptides. The material achieved enrichment through a dual-mode mechanism based on electrostatic and hydrophilic properties. The study successfully identified 2928 glycopeptides and 3051 phosphopeptides from mouse lung tissue samples, demonstrating the utility of this material for large-scale PTM analysis.

Simultaneous Profiling of Palmitoylomics and Glycomics with Photo/pH Dual-Responsive Magnetic Nanocomposites

Haijiao Zheng et al.

Summary: In this study, a novel dual-responsive magnetic nanocomposite was developed for the simultaneous enrichment of palmitoyl and glycopeptides. The nanocomposite exhibited flexible photo-, pH-, and magnetic-responsive capabilities, enabling the simultaneous recognition of hydrophobic palmitoyl peptides and hydrophilic glycopeptides. This platform provides a highly specific method for palmitoylomics and glycomics analysis, and has potential implications in clinical applications.

Boost-DiLeu: Enhanced Isobaric N,N-Dimethyl Leucine Tagging Strategy for a Comprehensive Quantitative Glycoproteomic Analysis

Danqing Wang et al.

Summary: An improved isobaric labeling strategy, Boost-DiLeu, was developed to enhance the quantitative glycoproteomic analysis, allowing reliable quantification of N-glycopeptides even from limited samples. The strategy was applied to differentiate N-glycosylation profiles between Alzheimer's disease patients and non-AD donors, revealing dysregulated processes and pathways affected by N-glycosylation in AD pathogenesis. The study demonstrated the feasibility of using this strategy for in-depth glycoproteomic analysis of size-limited clinical samples, showing promise for identifying novel therapeutic targets or biomarkers in systems with limited sample quantity.

Dual-Functional Ti(IV)-IMAC Material Enables Simultaneous Enrichment and Separation of Diverse Glycopeptides and Phosphopeptides

Junfeng Huang et al.

Summary: A new approach utilizing dual-functional Ti(IV)-IMAC material enables simultaneous enrichment and separation of glycopeptides, phosphopeptides, and M6P glycopeptides, as well as separation of neutral and sialyl glycopeptides and mono- and multi-phosphopeptides through different elution processes. This strategy significantly improves the coverage and identification numbers of these PTMs compared to conventional methods, providing novel insights into potential crosstalk among them.
