Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Role of nutraceuticals on neurodegenerative diseases: neuroprotective and immunomodulant activity

Angelo Fumia et al.

Summary: Neurodegeneration is a degenerative process characterized by the progressive loss of neuron structure and function, leading to dementia and other neurodegenerative diseases. By 2040, it is estimated that approximately 81.1 million people will be suffering from these diseases. In addition to traditional pharmacological therapy, natural nutraceuticals have played an important role in counteracting these diseases through their broad spectrum of biological effects.

Cross-seeding between Aβ and SEVI indicates a pathogenic link and gender difference between alzheimer diseases and AIDS

Yijing Tang et al.

Summary: This study reveals a potential crosstalk between Alzheimer's Disease (AD) and HIV/AIDS through the cross-seeding interactions between Aβ and SEVI proteins. The cross-seeding of SEVI with Aβ can prevent Aβ aggregation, reduce Aβ-induced cell toxicity, and attenuate Aβ-accumulated paralysis in transgenic AD C. elegans.

Atrial Natriuretic Peptide Associated with Cardiovascular Diseases Inhibits Amyloid-β Aggregation via Cross-Seeding

Yijing Tang et al.

Summary: Cardiovascular diseases (CVDs) and Alzheimer’s disease (AD) share common risk factors and have overlapping pathogenesis. This study explores the cross-seeding between CVD-associated atrial natriuretic peptide (ANP) and AD-associated fibrin amyloid (Afi) at the protein level. The results show that ANP can cross-seed with different forms of Afi and modify its aggregation pathway, fibril morphology, and cytotoxicity. Additionally, ANP can effectively inhibit Afi-induced neurodevelopmental toxicity in transgenic worms.

Repurposing of intestinal defensins as multi-target, dual-function amyloid inhibitors via cross-seeding

Yijing Tang et al.

Summary: This study proposes a new strategy for combating amyloid formation and microbial infection by repurposing intestinal defensins as amyloid inhibitors with dual functions. The intestinal defensins can cross-seed with amyloid peptides to prevent aggregation and also retain antimicrobial activity.

Cancer and Alzheimer's disease inverse relationship: an age-associated diverging derailment of shared pathways

Cristina Lanni et al.

Summary: Several epidemiological studies have shown a negative correlation between cancer and Alzheimer's disease. The review focuses on analyzing common biological mechanisms for both diseases, particularly highlighting the roles of PIN1 and p53 in understanding this inverse correlation. These molecular players may provide insight into potential therapeutic targets for both cancer and AD.

Summary: Genetic alterations, particularly the K-Ras mutation, play a major role in the progression of pancreatic precursor lesions into PDAC. The tumor microenvironment and immune evasion are challenges in therapeutic approaches, with targeting signaling molecules and immune checkpoint inhibition showing promising results in pancreatic cancer treatment.

Engineering Amyloid Aggregation as a New Way to Eliminate Cancer Stem Cells by the Disruption of Iron Homeostasis

Hongshuang Qin et al.

Summary: Recent research has shown that oligomeric Aβ1-42 and Aβ1-40 can suppress the viability of breast cancer stem cells by disrupting iron homeostasis, leading to iron accumulation in lysosomes, triggering ROS production, and inducing stem cell death. In vivo experiments demonstrate that pretreatment with OAβ1-42 inhibits tumor formation in breast cancer stem cells. These findings suggest that OAβ1-42 and OAβ1-40 have multifaceted capabilities in eliminating cancer stem cells.

Antimicrobial α-defensins as multi-target inhibitors against amyloid formation and microbial infection

Yanxian Zhang et al.

Summary: The study proposed a novel anti-amyloid and antimicrobial hypothesis using two host-defense antimicrobial peptides containing beta-rich structures to inhibit amyloid aggregation and microbial infection while retaining antimicrobial activity. The discovery of these peptides greatly expands the therapeutic potential of antimicrobial peptides as multi-target amyloid inhibitors for better understanding and treating amyloid diseases.
