Silver and copper-benznidazole derivatives as potential antiparasitic metallodrugs: Synthesis, characterization, and biological evaluation

Cassiano Cunha de Souza et al.

Summary: Four novel metal-BZN complexes were synthesized and characterized in this study. The IC50 values of these complexes in inhibiting parasite proliferation stages are five to ten times lower than benznidazole itself. The cytotoxicity in human cells is lower for these complexes compared to BZN, indicating higher selectivity.

Copper(II) and silver(I)-1,10-phenanthroline-5,6-dione complexes interact with double-stranded DNA: further evidence of their apparent multi-modal activity towards Pseudomonas aeruginosa

Anna Clara Milesi Galdino et al.

Summary: The study found that Cu-phendione has a higher DNA binding affinity, can induce DNA damage, and cause DNA fragmentation. These results suggest that complexes containing phendione may elicit toxicity toward the multidrug-resistant pathogen P. aeruginosa through a novel DNA-targeted mechanism.

Promising fluconazole based zinc(II) and copper(II) coordination polymers against Chagas disease

Jose Aleixo De Azevedo-Franca et al.

Summary: A series of new transition metal coordination polymers were synthesized and their structures were elucidated. These coordination polymers exhibited promising anti-parasitic activity, surpassing the free azole drug. Therefore, they are potential candidates for the treatment of Chagas disease.

Monotherapy and combination chemotherapy for Chagas disease treatment: a systematic review of clinical efficacy and safety based on randomized controlled trials

Silas Santana Nogueira et al.

Summary: This article systematically reviewed the clinical evidence on monotherapy and combination chemotherapy for the treatment of Chagas disease (ChD). The study found that benznidazole (BNZ) is the most viable option for treatment, but new protocols need to be developed to mitigate side effects and increase patient adherence.

Chagas Disease: Chronic Chagas Cardiomyopathy

Natalia Giraldo Echavarria et al.

Summary: Chagas disease is a tropical vector-borne infection caused by the parasite Trypanosoma cruzi, endemic in South and Central America but increasingly becoming a burden in the United States and Europe due to human migration. Most patients are asymptomatic during the acute phase, with around 30% progressing to detectable organ damage mainly affecting the cardiovascular and digestive systems in the chronic symptomatic phase, which can lead to Chagas cardiomyopathy, a leading cause of nonischemic cardiomyopathy in Latin America. Treatment during the parasitic infection is possible, but efficacy during the advanced chronic symptomatic phase remains uncertain. Further studies are needed to improve clinical outcomes and refine treatment recommendations.

Mechanisms Associated with Trypanosoma cruzi Host Target Cell Adhesion, Recognition and Internalization

Oscar Hernan Rodriguez-Bejarano et al.

Summary: Chagas disease, caused by Trypanosoma cruzi parasite, is mainly transmitted by insect bites. The parasite's lifecycle includes an intracellular phase and infective forms, involving various signaling cascades and intracellular associations for recognition and internalization in host cells.

Chagas Disease Chemotherapy: What Do We Know So Far?

Aline A. Zuma et al.

Summary: Chagas disease is a Neglected Tropical Disease affecting around 6-7 million people worldwide, predominantly in Latin America. Current treatments, benznidazole and nifurtimox, have limited effectiveness and can cause side effects, especially in the chronic phase. Studies have shown potential new drugs with equivalent or superior efficacy to benznidazole, highlighting the need for further research to improve treatment options.
