Directed evolution of the fusion enzyme for improving astaxanthin biosynthesis in Saccharomyces cerevisiae

Yong-Wen Ding et al.

Summary: This study improved the biosynthesis pathway of astaxanthin in yeast by utilizing directed evolution. By reducing the accumulation of intermediate products and increasing the content of astaxanthin, the fusion enzyme of crtZ-crtW with a 2 X GGGGS peptides linker showed promising results. Through directed evolution, 9 variant fusion mutants with improved astaxanthin production were generated, and a variant, L95S + I206L, was identified to have a significantly increased astaxanthin content compared to the control strain. Structural analysis suggested that the mutations altered the interaction between the substrate and the enzyme's pocket. These findings provide an efficient strategy to reduce the accumulation of intermediate products in complex biosynthesis pathways.

Metabolic engineering of Escherichia coli with electron channelling for the production of natural products

Seon Young Park et al.

Summary: This study reports an electron channelling strategy based on the application of Photorhabdus luminescens CipB scaffold protein, which allows efficient electron transfer between eukaryotic cytochrome P450s (P450s) and P450 reductase in bacteria. Using this strategy, recombinant Escherichia coli strains producing various natural products were developed and the production titres were enhanced through optimization of the culture conditions.

Metabolic Engineering of Non-carotenoid-Producing Yeast Yarrowia lipolytica for the Biosynthesis of Zeaxanthin

Yuxiao Xie et al.

Summary: This study constructed the β-carotene biosynthesis pathway in Yarrowia lipolytica and successfully produced zeaxanthin by introducing exogenous crtZ genes. The crtZ gene from Pantoea ananatis showed the best performance for zeaxanthin production, leading to significantly increased zeaxanthin titer through high-copy-number integration. The results demonstrated the potential of Y. lipolytica as a cell factory for zeaxanthin biosynthesis.

Biochemical and Immunological implications of Lutein and Zeaxanthin

Javaria Zafar et al.

Summary: Nature has been a key source of bioactive molecules throughout history, with human macular carotenoids attracting increasing attention. Lutein, zeaxanthin, and their metabolites are selectively accumulated in the macular region, but extracting them from natural sources like plants is costly and challenging. Microbial and microalgal carotenoids are considered as promising alternatives, with genetic engineering techniques like CRISPR/Cas9 enabling overproduction, although further research is needed for commercial applications. Esterification and microencapsulation can enhance the stability of these carotenoids and expand their applications in the food industry, especially for their antioxidant properties in various diseases.
