Anticancer therapeutic potential of benzofuran scaffolds

Summary: The benzofuran moiety is a key component in numerous biologically active natural and synthetic heterocycles. These heterocycles exhibit unique therapeutic potential and are implicated in various clinical drugs. Recent studies have confirmed the exceptional inhibitory potency of benzofurans against human cancer cell lines compared to reference anticancer drugs. Several publications have focused on the anticancer properties of benzofuran-based heterocycles. This review comprehensively covers the latest developments in the anticancer activities of natural and synthetic benzofuran scaffolds from 2019 to 2022, highlighting their potential as promising candidates for anticancer agents.

Intermolecular Interactions of 3,5-bis(4-Methoxyphenyl)-4,5-dihydro-1H-pyrazole-1-carbothioamide in a Cocrystal with 1,3-bis(4-Methoxyphenyl)prop-2-en-1-one and Dimethylformamide Solvate

Summary: Two new multicomponent crystals consisting of 3,5-bis(4-methoxyphenyl)-4,5-dihydro-1H-pyrazole-1-carbothioamide (1) with 1,3-bis(4-methoxyphenyl)prop-2-en-1-one (2) and with dimethylformamide (DMF) were prepared and structurally characterized. The comparison of hydrogen bonding contacts between different crystal structures of 1 revealed variations in the orientation of the methoxy groups and the combinations of hydrogen bonding contacts. The involvement of different atoms in the hydrogen bonding contacts also differed between the pure compound and the multicomponent crystals.

Antifungal Thiazolidines: Synthesis and Biological Evaluation of Mycosidine Congeners

Summary: Novel derivatives of Mycosidine were synthesized through Knoevenagel condensation and reactions with thiazolidines, showing high antifungal activity with morphological changes in Candida yeast cell wall. Limited proteomic screening and toxicity analysis in mutants indicate that Mycosidine activity is linked to glucose transport, suggesting a novel mechanism of action for this first-in-class antifungal drug.

Summary: This study reported the synthesis of chalcones by reacting 1-(5-methyl)-1H-1,2,3-triazol-4-yl)ethan-1-ones with benzaldehydes under basic conditions in ethanol. Furthermore, it was found that the synthesized chalcones could react with thiosemicarbazide, followed by reaction with specific ketones, to yield pyrazolyl-thiazole compounds with novel structures. The structures of six newly synthesized heterocycles were confirmed by X-ray crystallography.

Summary: Reaction of 1-(5-methyl-1-(4-nitrophenyl)-1H-1,2,3-triazol-4-yl)ethan-1-one (1) and N-phenylhydrazinecarbothioamide (2) in boiling ethanol with a catalytic amount of concentrated hydrochloric acid yielded (Z)-2-(1-(5-methyl-1-(4-nitrophenyl)-1H-1,2,3-triazol-4-yl)ethylidene)-N-phenylhydrazine-1-carbothioamide (3) with an 88% yield. The structure of compound 3 was confirmed by single-crystal X-ray diffraction and magnetic resonance spectroscopy.
