Epithelial-Mesenchymal Transition (EMT) as a Therapeutic Target

Summary: Metastasis, the spread of cancer cells to distant sites, is the primary cause of cancer morbidity and mortality. During the initial stages of metastasis, cancer cells undergo EMT, becoming migratory and invasive mesenchymal-like cells with stem cell properties. Targeting EMT in therapy has the potential to prevent cancer cell invasion and dissemination, reduce cancer stemness, and enhance the effectiveness of chemotherapy.

Assessing the role for nailfold videocapillaroscopy in interstitial lung disease classfication: a systematic review and meta-analysis

Summary: This study conducted a meta-analysis on the efficacy of nailfold videocapillaroscopy (NVC) in subgroups of connective tissue disease-related interstitial lung disease (CTD-ILD), interstitial pneumonia with autoimmune features (IPAF), and idiopathic pulmonary fibrosis (IPF). The results showed that NVC can increase the diagnostic accuracy of ILD when used in a multi-disciplinary setting, with the highest utility in CTD-ILD, followed by IPAF and IPF. The late scleroderma pattern was found to be the most frequent nailfold capillary pattern.

Evidence of telomere attrition and a potential role for DNA damage in systemic sclerosis

Summary: Cell senescence is associated with telomere shortening in systemic sclerosis (SSc), especially in patients with lung disease or anti-topoisomerase I antibodies. Targeting DNA damage with the GSE4 peptide attenuates experimental skin fibrosis and reduces profibrotic factor expression in fibroblasts, suggesting a role for oxidative DNA damage in scleroderma.

Many studies have found higher levels of certain cytokines/chemokines in BAL from SSc patients with impaired lung function and lung involvement.

Lung biopsy may not be necessary for confirming the presence of interstitial lung disease in the right clinical setting, with surgical lung biopsies often reserved for atypical presentations. This review discusses histological findings in SSc-ILD and other changes sometimes seen in lung biopsies from systemic sclerosis patients.
