miR-16-5p Suppression Protects Human Cardiomyocytes against Endoplasmic Reticulum and Oxidative Stress-Induced Injury

Summary: Oxidative stress plays a significant role in cardiovascular diseases, and endoplasmic reticulum stress is an important source of reactive oxygen species. miR-16-5p could promote oxidative stress through ER stress induction and regulation of ATF6. Analysis of oxidative markers in the plasma of ischemic dilated cardiomyopathy patients showed a correlation with oxidative stress. Overexpression of miR-16-5p increased oxidative stress in cardiomyoblasts, while its suppression decreased apoptosis, inflammation, and cardiac damage through activation of the ATF6-mediated cytoprotective pathway. ATF6 was identified as a direct target gene of miR-16-5p.

The results showed that overexpression of TSP1 activated endoplasmic reticulum stress through the ATF6-CHOP axis, inhibited cell proliferation, and promoted apoptosis and endoplasmic reticulum stress in HK-2 cells.
