Application of in vitro PAMPA technique and in silico computational methods for blood-brain barrier permeability prediction of novel CNS drug candidates

Summary: Assessing the permeability of small molecules through the blood-brain barrier is important for the development of CNS drug candidates. In this study, in silico and in vitro methods were used to predict the permeability of compounds affecting serotonin and dopamine levels in the CNS. A reliable QSPR model was created, which can be used as a pre-screening tool for predicting the BBB permeability of structurally related CNS compounds.

Summary: This study investigates the performance variation of three docking algorithms, GOLD, Glide, and PLANTS, in replicating the coordinates of crystallographic ligands of SARS-CoV-2 main protease. The importance of solvent exposure in docking performance is highlighted through the comparison of docking data and values derived from solvent exposure calculations. Molecular dynamics simulations further confirm that protein-ligand complexes with higher solvent exposure tend to be less stable.

QSAR, ADMET In Silico Pharmacokinetics, Molecular Docking and Molecular Dynamics Studies of Novel Bicyclo (Aryl Methyl) Benzamides as Potent GlyT1 Inhibitors for the Treatment of Schizophrenia

Summary: Forty-four bicyclo((aryl)methyl)benzamides were developed as inhibitors of glycine transporter type 1 (GlyT1) using molecular modeling techniques. QSAR models confirmed a strong correlation between the biological inhibitory activity against schizophrenia and physicochemical, geometrical, and topological descriptors. In silico ADMET properties indicated that the most active ligands have a high probability of penetrating the central nervous system (CNS), while one molecule showed a higher probability of being absorbed by the gastrointestinal tract. Molecular docking and dynamics simulations revealed the active sites and stability of the ligands in the dopamine transporter (DAT) protein, suggesting their potential as candidate drugs for schizophrenia treatment.

Multivariate Analysis Applied to Aquifer Hydrogeochemical Evaluation: A Case Study in the Coastal Significant Subterranean Water Body between Cecina River and San Vincenzo, Tuscany (Italy)

Summary: The hydrogeochemical characteristics of a significant subterranean water body in Italy were evaluated using multivariate statistical analysis methods, revealing that major ions had a significant influence over groundwater chemistry. Three clusters were identified based on the chemical characteristics of the water samples: low salinity/mixed waters, high salinity waters, and low salinity/fresh waters. Temporal changes in cluster assignment were observed in only two wells, indicating the impact of hydrodynamic conditions on groundwater chemistry. The results were also displayed using the classical hydrochemical approach of the Piper plot, showing the effectiveness of this method in quickly analyzing large datasets and identifying spatiotemporal patterns.
