Design, synthesis, and SAR studies of novel 4-methoxyphenyl pyrazole and pyrimidine derivatives as potential dual tyrosine kinase inhibitors targeting both EGFR and VEGFR-2

Summary: By guided by the pharmacophoric features of EGFR and VEGFR-2 antagonists, two series of novel compounds were designed and synthesized as dual EGFR/VEGFR-2 inhibitors. Compound 12 showed strong antiproliferative effects and high inhibitory activities against both EGFR and VEGFR-2, making it a potential drug candidate. Further evaluation and computational studies revealed promising results and provided insights for designing more potent drug candidates.

Anticoagulants as Potential SARS-CoV-2 Mpro Inhibitors for COVID-19 Patients: In Vitro, Molecular Docking, Molecular Dynamics, DFT, and SAR Studies

Summary: In this study, 34 anticoagulant drugs were screened in silico against the main protease (M-pro) of SARS-CoV-2 using molecular docking tools. The most promising drugs, including Idraparinux, fondaparinux, eptifibatide, heparin, and ticagrelor, demonstrated high binding affinities towards SARS-CoV-2 M-pro. Molecular dynamics and quantum mechanical studies were conducted to provide further insights into the properties of these drugs. Biological evaluations were performed using in vitro assays, and it was found that ticagrelor exhibited the highest inhibitory potential against SARS-CoV-2.
