Effect of matrix heterogeneity on cell mechanosensing

Copper-Free Azide-Alkyne Cycloaddition for Peptide Modification of Alginate Hydrogels

Summary: Alginate, a biocompatible polymer derived from algae, is often used in tissue engineering as a synthetic version of the extracellular matrix. This study developed a copper-free, strain-promoted azide-alkyne cycloaddition (SPAAC)-mediated method to conjugate azide-modified adhesive peptides (Az-cRGD) onto alginate, improving peptide bioactivity and enhancing osteogenesis in 3D alginate gels. This approach overcomes limitations of previous bioconjugation methods and has the potential to expand ligand possibilities for tissue engineering applications.
