Extracellular vesicle-loaded hydrogels for tissue repair and regeneration

Yikun Ju et al.

Summary: Extracellular vesicles (EVs) are secreted by cells and have significant potential in regenerative medicine. Mesenchymal stem cells can secrete EVs that promote tissue repair and regeneration. However, EVs are rapidly cleared by the circulatory system, so using hydrogels to carry and slow the release of EVs can be an effective approach.

Proteomic analysis of extracellular vesicles secreted by primary human epithelial endometrial cells reveals key proteins related to embryo implantation

Marina Segura-Benitez et al.

Summary: Successful implantation relies on communication between maternal endometrium and embryo, with EVs playing a role in this process. Ultracentrifugation was found to be the most efficient method for EV isolation from endometrial epithelial cells, with EVs containing proteins related to embryo implantation processes.

Exporting Proteins Associated with Senescence Repair via Extracellular Vesicles May Be Associated with Early Pregnancy Loss

Yi Zhang et al.

Summary: In this study, the protein profiles in placental extracellular vesicles (EVs) derived from missed miscarriage were compared with those from healthy pregnancy. The results showed abundant proteins associated with DNA damage/repair and cell death/survival in placental EVs from missed miscarriage. Additionally, the levels of three senescence-repair-associated proteins (RPA-70, PMSE-4, and PAK-2) were found to be lower in missed-miscarriage placentae but higher in GW4869-treated placental explants. These findings suggest that dysfunction of placental development in missed miscarriage may be associated with inadvertently sorted cargos and exported proteins by placental EVs.

Summary: Macrophages are crucial components of the innate immune system and play a significant role in pregnancy by contributing to various processes that support reproductive success. Disruption of the local tissue microenvironment can affect macrophage function and is linked to adverse obstetric outcomes. Understanding the environmental influences on macrophage polarization at the maternal-fetal interface could help in alleviating adverse pregnancy outcomes.

Summary: Research has shown that EVs released from the male reproductive tract play a crucial role in the development and function of sperm by carrying RNA, protein, and lipid cargoes for post-testicular modification and development of sperm. While little is known about the interaction between EVs from the testes and the spermatogonial stem cell niche or developing sperm within the seminiferous tubules, these interactions are vital for the subsequent function of sperm.

Protein Fingerprinting of Seminal Plasma Reveals Dysregulation of Exosome-Associated Proteins in Infertile Men with Unilateral Varicocele

Summary: This study identified differentially expressed proteins in seminal plasma of unilateral varicocele patients compared to fertile donors, with significant upregulation of ANXA2 and downregulation of KIF5B. The main upstream regulators of these proteins in unilateral varicocele group were androgen receptor, YB1, and NRF2. KIF5B and ANXA2 could potentially serve as biomarkers for infertility in unilateral varicocele patients.
