Degradation and biological performance of porous osteomimetic biphasic calcium phosphate in vitro and in vivo

Summary: The combination of biphasic calcium phosphate (BCP) with osteomimetic porous micro-structure obtained via freeze casting is believed to achieve excellent bone regeneration, while the effects of different ratios of HA and beta-TCP on the degradation and biological performance of the BCP scaffolds need to be studied. The BCP scaffolds with varying HA/beta-TCP ratios demonstrated similar microstructure, mechanical performance, and protein absorption capacity, with the HA70/beta-TCP30 ratio showing optimal degradation rate. The in vitro and in vivo experiments confirmed the biocompatibility and bone regeneration capacity of the porous osteomimetic BCP scaffold with a 30:70 HA/beta-TCP ratio.

Nanoscale Zeolitic Imidazolate Framework-8 Activator of Canonical MAPK Signaling for Bone Repair

Summary: This study demonstrates that nano ZIF-8 can drive rat bone mesenchymal stem cells differentiation into osteoblasts, with better osteogenic effect compared to ionic conditions of Zn at the same concentration. The cellular uptake mechanisms were revealed to involve caveolae-mediated endocytosis and macropinocytosis. Additionally, transcriptome sequencing identified upregulation of multiple osteogenic pathways, primarily phosphorylating ERK to activate the canonical mitogen-activated protein kinase pathway for promoting osteogenesis.
