Studying Cardiac Neural Network Dynamics: Challenges and Opportunities for Scientific Computing

Nil Z. Gurel et al.

Summary: The neural control of the heart involves continuous regulation of cardiac activity to meet the body's need for blood flow. This regulation is achieved through interconnected neural networks that work together with central and peripheral components. Computational methods can improve experimental design and help understand how information is processed during closed-loop control. However, processing and analyzing the large cardio-neural datasets generated by these experimental designs present computational challenges in terms of data sharing and reproducibility. Overcoming these challenges can enhance our understanding of cardiac pathologies and contribute to clinical implementation.

Critical repolarization gradients determine the induction of reentry-based torsades de pointes arrhythmia in models of long QT syndrome

Mathilde R. Rivaud et al.

Summary: The study found that torsades de pointes arrhythmia can be induced within a critical range of repolarization time and repolarization time heterogeneities, maintained by reentry wandering along the repolarization gradient. Local restitution curves from the long and short repolarization time regions crossed, associated with T wave inversion at coupling intervals at either side of the crossing point.

Long-term Follow-Up of Patients with Heart Failure and Reduced Ejection Fraction Receiving Autonomic Regulation Therapy in the ANTHEM-HF Pilot Study

Kamal Sharma et al.

Summary: The ANTHEM-HF pilot study showed that autonomic regulation therapy (ART) utilizing cervical vagus nerve stimulation (VNS) was associated with sustained improvements in left ventricular function and heart failure symptoms at 6 and 12 months. Long-term follow-up of 42 months demonstrated that ART was durable, safe, and had beneficial effects on LVEF and 6-min walk distance in patients with HFrEF. This suggests that chronic, open-loop ART continued to be well-tolerated in this population, with further evaluation underway in the ANTHEM-HFrEF Pivotal Study.

Single-cell transcriptomic profiling of satellite glial cells in stellate ganglia reveals developmental and functional axial dynamics

Valerie Y. H. van Weperen et al.

Summary: Single-cell RNA sequencing of mouse stellate ganglia identified six distinct transcriptomic subtypes of satellite glial cells (SGCs), with one subtype characterized by pro-inflammatory markers. Over maturation or differentiation axes, SGCs showed gradual shifts in developmental and functional pathways, suggesting dynamic physiological adaptations that modulate neuronal function. Unbiased pseudo-time analysis revealed two distinct trajectories involving the studied SGC subtypes, highlighting the heterogeneity and dynamic nature of SGCs in modulating neurotransmission.

A novel metric linking stellate ganglion neuronal population dynamics to cardiopulmonary physiology

Koustubh B. Sudarshan et al.

Summary: Stellate ganglion neuronal populations exhibit specificity and integration towards the near-peak systolic phase of the cardiac cycle, providing insight into cardiopulmonary function. This novel approach of linking neuronal activity to mechanical events during systole can be used to study stellate ganglion control in normal and disease states.
