Population pharmacokinetics of anidulafungin in ICU patients assessing inter- and intrasubject variability

Iasonas Kapralos et al.

Summary: The population pharmacokinetics of anidulafungin in critically ill patients in ICUs showed significant interindividual and interoccasion variability, with reduced probability of target attainment compared to healthy volunteers. Pharmacokinetic simulations demonstrated the impact of dosing regimens on target attainment probabilities.

Evaluation of Synergistic Activity of Isavuconazole or Voriconazole plus Anidulafungin and the Occurrence and Genetic Characterization of Candida auris Detected in a Surveillance Program

Michael A. Pfaller et al.

Summary: A total of 36 Candida auris isolates were evaluated for antifungal susceptibility and synergy against anidulafungin plus voriconazole or isavuconazole. The isolates showed resistance to multiple antifungal classes, with varying degrees of synergy observed with different combinations of antifungal drugs. Genetic analysis revealed isolates from different regions were genetically related but distinct. Erg11 alterations were associated with resistance to some antifungal agents.

Comparison of In Vitro Killing Activity of Rezafungin, Anidulafungin, Caspofungin, and Micafungin against Four Candida auris Clades in RPMI-1640 in the Absence and Presence of Human Serum

Renato Kovacs et al.

Summary: Candida auris is an emerging and frequently multidrug-resistant pathogen for which echinocandins are the preferred treatment option. The study compared the killing activities of four echinocandins against different clades of C. auris, with rezafungin showing the same or better activity at clinically attainable trough concentration against all four clades.

In Vitro Synergistic Interactions of Isavuconazole and Echinocandins against Candida auris

Unai Caballero et al.

Summary: Combining isavuconazole with echinocandins showed an overall synergistic interaction against Candida auris, proving to be more effective than monotherapy regimens. Synergism and fungistatic activity were achieved with combinations that included low concentrations of isavuconazole and high concentrations of echinocandins. Results indicated that increasing drug concentrations did not improve antifungal activity once synergy was achieved.

Unpredictable In Vitro Killing Activity of Amphotericin B against Four Candida auris Clades

Zoltan Papp et al.

Summary: The study found that AMB has weak killing activity against C. auris regardless of clades, even when MIC values are low (<= 1 mg/L). The killing activity of AMB is concentration-, clade-, and isolate-dependent, making its efficacy unpredictable in the treatment of invasive C. auris infections.
