Assessment of biomarkers of exposure and potential harm, and physiological and subjective health measures in exclusive users of nicotine pouches and current, former and never smokers

David Azzopardi et al.

Summary: Exclusive users of Velo NPs have significantly less exposure to tobacco toxicants and more favorable biomarkers associated with smoking-related diseases compared to smokers.

A Randomised Study to Investigate the Nicotine Pharmacokinetics of Oral Nicotine Pouches and a Combustible Cigarette

Michael McEwan et al.

Summary: This study compared the pharmacokinetics and subjective effects of nicotine absorption from different nicotine pouches and a combustible cigarette. The results showed that nicotine absorption from nicotine pouches can be similar to conventional cigarettes, but the subjective effects are not as strong as cigarettes. Further research is needed to understand how other physical characteristics of nicotine pouches may impact nicotine delivery, pharmacokinetics, and subjective responses.

Chemical characterization of tobacco-free modern oral nicotine pouches and their position on the toxicant and risk continuums

David Azzopardi et al.

Summary: Compared with snus, NPs have lower levels of harmful compounds and expose users to lower levels of toxic compounds. NPs and NRTs have the lowest toxicant profiles and estimations of relative toxicant exposure across the product categories.

A randomised, open-label, cross-over clinical study to evaluate the pharmacokinetic, pharmacodynamic and safety and tolerability profiles of tobacco-free oral nicotine pouches relative to cigarettes

Fiona Chapman et al.

Summary: This study compared the pharmacokinetic, pharmacodynamic, safety, and tolerability profiles of two nicotine pouch variants with cigarettes. The results demonstrated that nicotine pouches may provide an acceptable alternative for adult smokers to achieve satisfactory nicotine delivery levels with lower abuse liability and addictive potential compared to continued cigarette smoking.

Nicotine pharmacokinetics and subjective responses after using nicotine pouches with different nicotine levels compared to combustible cigarettes and moist smokeless tobacco in adult tobacco users

Jianmin Liu et al.

Summary: Oral tobacco-derived nicotine products, such as nicotine pouches (NPs), have the potential to be acceptable alternatives for cigarette and moist smokeless tobacco (MST) users. The abuse potential of NPs is not likely to be higher than cigarettes and MST.

Nicotine pharmacokinetics and subjective response among adult smokers using different flavors of on!® nicotine pouches compared to combustible cigarettes

Jesse Rensch et al.

Summary: Based on the nicotine PK parameters and subjective responses, we conclude that NPs, regardless of flavor, likely have lower abuse potential than cigarettes. Overall, this study suggests that the NPs may be potentially acceptable switching products for adult smokers.

Examining Market Trends in Smokeless Tobacco Sales in the United States: 2011-2019

Cristine D. Delnevo et al.

Summary: The smokeless tobacco industry in the U.S. has experienced various trends from 2011 to 2019, with moist snuff dominating the market but showing a recent decline, while snus sales have been consistently increasing. On the other hand, sales of chewing tobacco, dry snuff, and dissolvable products have been decreasing. Emerging tobacco-free nicotine products captured a share of the market since 2016.

A screening approach for the evaluation of tobacco-free 'modern oral' nicotine products using Real Time Cell Analysis

N. East et al.

Summary: The study proposes an application for the RTCA platform to provide a screening approach for evaluating the cytotoxic potential of tobacco-free nicotine pouches. Results show that H292 cells can accurately predict cytotoxicity outcomes and offer a more compatible test system for fast screening studies.
