Molybdenum Cofactor Deficiency in Humans

Summary: Molybdenum cofactor deficiency is a disease characterized by neonatal-onset myoclonic epileptic encephalopathy and dystonia, with similar cerebral MRI changes to hypoxic-ischemic lesions. The cause of the disease is the loss of sulfite oxidase activity, leading to the accumulation of toxic sulfite and disruption of metabolic processes. Different types of the disease are classified based on the impairment of specific synthetic steps, with type A being treatable with cyclic pyranopterin monophosphate. There is currently no specific treatment for type B/C and SOX deficiency, but recent progress has been made in understanding the underlying metabolic changes and proposing potential therapeutic interventions.
