Microfluidic aspiration-assisted electrical impedance spectroscopy system is a reliable tool for the characterization of oocyte hardening

Yuan Cao et al.

Summary: The fertilization success in assisted reproduction is heavily influenced by the quality of the oocyte, particularly the hardening state of the zona pellucida. Existing techniques for reproduction involve manual manipulation, which is expensive, non-standardized, and reliant on individual observers. To address this, a microfluidic-based cell trap technique was proposed, allowing for simple oocyte handling and measurement of zona pellucida aspiration via electrical impedance spectroscopy. This technique can be used for artificial fertilization in both human and animal reproductive medicine, as well as for the development of contraceptives and characterization of single cell rheological properties.

The E-modulus of the oocyte is a non-destructive measure of zona pellucida hardening

Carlo Schmitz et al.

Summary: The elastic modulus of the zona pellucida can reflect the cleavage of ZP2 in oocytes, providing a new method for assessing oocyte fertility with increased sensitivity. This measurement can define the range of zona hardening and could potentially serve as a non-destructive marker-free method for evaluating individual oocyte quality.

Dry Film Resist Laminated Microfluidic System for Electrical Impedance Measurements

Yuan Cao et al.

Summary: A three-level DFR lamination process using SUEX for a microfluidic chip with vertically arranged microelectrodes for electrical impedance measurements is presented. The setup traps and fixes objects under test to the electrodes using an aperture in the center of ring-shaped electrodes and a microfluidic suction channel. The electrical impedance measurements show higher values when spheres are trapped, with the magnitude depending on the sizes of the spheres.
