Evolving a Generalist Biosensor for Bicyclic Monoterpenes

Simon d'Oelsnitz et al.

Summary: Prokaryotic transcription factors have been repurposed as analytical and synthetic tools for precise chemical measurement and regulation. In this study, genetically encoded, generalist monoterpene biosensors were developed by using directed evolution to expand the effector specificity of the camphor-responsive TetR-family regulator CamR. These biosensors can recognize four distinct monoterpenes and have a high signal-to-noise ratio. They can serve as a starting point for the high-throughput screening and dynamic regulation of bicyclic monoterpene production strains.

Structural basis for the tryptophan sensitivity of TnaC-mediated ribosome stalling

Anne-Xander van der Stel et al.

Summary: The study identifies a mechanism where the TnaC-ribosome complex captures L-Trp to initiate termination arrest, with nascent TnaC preventing release factor 2 from adopting an active conformation. The relative rates of L-Trp binding and peptidyl-tRNA cleavage determine the tryptophan sensitivity of each variant. This reveals a strategy whereby a nascent peptide assists the ribosome in detecting a small metabolite.
