Cdc6 is sequentially regulated by PP2A-Cdc55, Cdc14, and Sic1 for origin licensing in S. cerevisiae

Jasmin Philip et al.

Summary: This study reveals that PP2A(Cdc55) dephosphorylates the N-terminal sites of Cdc6 to release Clb2, while Cdc14 dephosphorylates the C-terminal phospho-degron to stabilize Cdc6 in mitosis. Additionally, the Cdk1 inhibitor Sic1 releases Clb2·Cdk1·Cks1 from Cdc6 to load Mcm2-7 on the chromatin upon mitotic exit. The findings suggest that phosphatases promote pre-RC assembly and origin licensing through the attenuation of different Cdk1-dependent inhibitory mechanisms of Cdc6.

Spatiotemporal coordination of Greatwall-Endos-PP2A promotes mitotic progression

Myreille Larouche et al.

Summary: Mitotic entry involves the inhibition of PP2A-B55/Tws, which is induced by Greatwall phosphorylation of Endos. The correct localization and phosphorylation of Tws and Endos play essential roles in mitotic regulation. The spatiotemporal regulation of Endos by Greatwall is conserved in humans and disruptions lead to mitotic defects.

Bora phosphorylation substitutes in trans for T-loop phosphorylation in Aurora A to promote mitotic entry

N. Tavernier et al.

Summary: The research uncovers the activation mechanism of AURKA crucial for mitotic entry, showing that phosphorylated Bora directly activates AURKA kinase activity. Through specific amino acid regions, Bora binds AURKA and substitutes for a key phospho-regulatory site, highlighting the importance of this mechanism for timely mitotic entry in Xenopus and human cells.

Mitotic timing is differentially controlled by A- and B-type cyclins and by CDC6 associated with a bona fide CDK inhibitor Xic1 in Xenopus laevis cell-free extract

Mohammed El Dika et al.

Summary: This study demonstrates the differential regulation of mitotic kinase activity by cyclin A and B, as well as the critical role of CDC6 in this process. Experimental increases in cyclin A only modify the level of histone H1 kinase activity, while cyclin B increases affect both the activity level and the timing of full activation. Additionally, CDC6 inhibits cyclin B-associated kinase activity but does not affect mitotic timing without cyclin A. The study also suggests that Xic1 plays a crucial role in coordinating the progression of M-phase by interacting with mitotic CDK complexes.
