Inflammasome activation in infected macrophages drives COVID-19 pathology

Esen Sefik et al.

Summary: This study demonstrates that lung-resident human macrophages play a critical role in the development of lung inflammation caused by SARS-CoV-2 infection. Macrophages activate inflammasomes, release inflammatory cytokines, and undergo pyroptosis in response to infection, contributing to the hyperinflammatory state in the lungs. Inhibition of the NLRP3 inflammasome pathway reverses chronic lung pathology, indicating the importance of inflammasome activation in lung inflammation.

Mechanism of filament formation in UPA-promoted CARD8 and NLRP1 inflammasomes

L. Robert Hollingsworth et al.

Summary: NLRP1 and CARD8 form supramolecular signaling complexes called canonical inflammasomes upon activation, leading to caspase-1 activation, cytokine maturation, and/or pyroptotic cell death. Their C-terminal fragments containing a CARD and the UPA subdomain facilitate self-oligomerization, recruiting the inflammasome adaptor ASC or caspase-1 for further signaling.

Structural basis for distinct inflammasome complex assembly by human NLRP1 and CARD8

Qin Gong et al.

Summary: NLRP1 and CARD8 are two recently characterized sensor proteins for the human inflammasome complex. The cryo-EM CARD filament structures of the NLRP1 and CARD8 activating domains reveal how NLRP1 and CARD8 discriminate between ASC and pro-caspase-1. The authors propose a two-step model for NLRP1 activation.

Distinct axial and lateral interactions within homologous filaments dictate the signaling specificity and order of the AIM2-ASC inflammasome

Mariusz Matyszewski et al.

Summary: Inflammasomes, such as AIM2-ASC, are essential filamentous signaling platforms in innate immunity. The similarity in structure between AIM2 and ASC filaments have been revealed through cryo-EM and simulations methods, shedding light on the assembly and recognition mechanisms of these filaments.

The AIM2 and NLRP3 inflammasomes trigger IL-1-mediated antitumor effects during radiation

Chuanhui Han et al.

Summary: Research showed that tumors lacking certain molecules in the inflammasome pathway were sensitive to radiation treatment in mice, while dual deficiency in multiple molecules resulted in radioresistance. The study also demonstrated that activation of AIM2 and NLRP3 inflammasomes by irradiated tumors played a role in inducing antitumor effects through IL-1 signaling in dendritic cells.

Computationally-guided design and affinity improvement of a protein binder targeting a specific site on HER2

Tae Yoon Kim et al.

Summary: This study describes the computationally-guided design and affinity improvement of a protein binder recognizing a specific site on domain IV of HER2. The binding affinity of the selected lead was significantly increased through mutual feedback between computational and experimental methods. The utility and potential of this approach was demonstrated by determining the binding interface of the developed protein binder through crystal structure analysis.
