Quantitative phosphoproteomics reveals ectopic ATP synthase on mesenchymal stem cells to promote tumor progression via ERK/c-Fos pathway activation

Summary: The tumor microenvironment plays a crucial role in cancer development. This study reveals that mesenchymal stem cells (MSCs), a key regulator of the microenvironment, promote lung cancer progression through the ERK/phospho-c-Fos-S374 pathway, as demonstrated by quantitative phosphoproteomics analysis. Additionally, the study also highlights the role of extracellular ATP, secreted by ATP synthase on the surface of MSCs, in activating the ERK/phospho-c-Fos-S374 pathway in lung cancer cells. These findings contribute to a better understanding of the complex interactions within the tumor microenvironment and provide potential targets for therapeutic interventions.

Summary: The study characterized the interactions between Penaeus monodon ATPsyn-beta and WSSV structural proteins, identifying VP51B and VP150 as proteins able to interact with PmATPsyn-beta. The results demonstrated that VP51B and VP150 are envelope proteins with portions exposed outside the WSSV virion, and they co-localize with PmATPsyn-beta on cell membranes. In vivo neutralization assays showed that recombinant VP51B and VP150 could delay mortality in shrimp challenged with WSSV.
