Antibodies gone bad - the molecular mechanism of light chain amyloidosis

Pharmacologic targeting of plasma cell endoplasmic reticulum proteostasis to reduce amyloidogenic light chain secretion

Summary: Current treatments for AL involve chemotherapeutics targeting the plasma cell population, but do not directly reduce toxic LC aggregation. A new strategy involves enhancing ER proteostasis to reduce amyloidogenic LC secretion, and compound 147 has been shown to reduce secretion of toxic LCs in AL patient-derived plasma cells through selective covalent modification of ER proteostasis factors. This approach could be used in combination with chemotherapeutics to reduce LC toxicity associated with AL pathogenesis.
