Single-particle combinatorial multiplexed liposome fusion mediated by DNA

Summary: In this study, a single-particle combinatorial multiplexed liposome fusion mediated by DNA is presented, enabling multistep and non-deterministic fusion of nanocontainers. Efficient and leakage-free stochastic fusion sequences were observed for arrays of target liposomes functionalized with individual DNA strands and labeled with distinct chromophores. The method allows high-throughput combinatorial multiplex screens using minimal material.

An Aldehyde Responsive, Cleavable Linker for Glucose Responsive Insulins

Summary: A glucose responsive insulin (GRI) has been developed using glucose cleavable linkers based on hydrazone and thiazolidine structures, demonstrating glucose responsiveness in vitro, particularly for thiazolidines. Clamp studies confirmed increased glucose infusion at hyperglycemic conditions for one GRI, indicating a true glucose response. The glucose responsive cleavable linker in these GRIs drives the release of active insulin from a circulating depot in response to changes in glucose levels.

Single-Molecule Study of Thermomyces lanuginosus Lipase in a Detergency Application System Reveals Diffusion Pattern Remodeling by Surfactants and Calcium

Summary: This study utilized single-molecule tracking to observe the real-time spatial localization and diffusional behavior of Thermomyces lanuginosus lipase (TLL) enzymes in a detergency application system. By imaging thousands of individual enzymes, the researchers directly observed and quantified the abundance and interconversion kinetics between three diffusional states that were found to correlate with function. The results showed how detergency effectors can influence the sampling of lipase functional states and alter protein functional dynamics and surface binding, ultimately affecting enzyme performance.

Interactions of Cell-Penetrating Peptide-Modified Nanoparticles with Cells Evaluated Using Single Particle Tracking
