Spontaneous tumor regression mediated by human T cells in a humanized immune system mouse model

Summary: Immunodeficient mice reconstituted with a human immune system (HIS mice) are a valuable tool for studying human immune responses to tumors. However, these mice often exhibit sub-optimal immune responses and lack antigen-specific B or T cell memory. In this study, we observed spontaneous regression of human B cell lymphoma Raji in HIS mice. Tumor regression was dependent on CD4+ and CD8+ T cells and resulted in T cell memory. We also found clonal expansion of specific T cell receptors (TCRs) and Raji-specific T cell reactivity in regressing tumors.

Summary: This study systematically characterizes the molecular signatures of interactions between T cells and antigen-presenting myeloid cells in the tumor microenvironment. The authors identify CD4(+)PD-1(+)CXCL13(+) T cells as a major interacting hub with antigen-presenting cells in human non-small cell lung carcinoma. They define this clonally expanded, tumor-specific and conserved T-cell subset as T-helper tumor (Tht) cells. The findings suggest that modulating Tht-dendritic cell interaction checkpoints could be a major strategy in immunotherapy.

Ex vivo confirmation of the cytolytic tumor-specific CD4 T cells was achieved using peptide-MHCII-multimer technology. The cytotoxic activity of these cells, partially dependent on SLAMF7, was demonstrated to have delayed kinetics compared to classical cytotoxic lymphocytes, and agonistic engagement of SLAMF7 enhanced their cytotoxicity, indicating potential synergy with other cancer immunotherapies.

Monocytes activated by toll-like receptor 4 agonists efficiently trigger the expansion of Th-CXCL13 cells, with TGF-beta 1 stimulation and activation of Sox4 being crucial for their induction and polarization. Additionally, the recruitment of TABs by Th-CXCL13 cells in TLSs induces plasma cell differentiation and immunoglobulin production, potentially leading to improved survival.
