Lipoprotein(a): A Genetically Determined, Causal, and Prevalent Risk Factor for Atherosclerotic Cardiovascular Disease: A Scientific Statement From the American Heart Association

Gissette Reyes-Soffer et al.

Summary: High levels of Lp(a) are an independent and causal risk factor for atherosclerotic cardiovascular diseases. Genetic factors determine 70% to 90% of interindividual heterogeneity in Lp(a) levels. There is a lack of standardized assays, diagnostic guidelines, and targeted treatments for lowering Lp(a). Understanding the genetic and biological basis for variation in Lp(a) levels in different populations is crucial.

Sortilin restricts secretion of apolipoprotein B-100 by hepatocytes under stressed but not basal conditions

Donna M. Conlon et al.

Summary: Genetic variants at the SORT1 locus in humans are associated with reduced plasma levels of LDL cholesterol and apoB. Loss of Sort1 expression in mice leads to increased apoB secretion under lipid loading or ER stress conditions. Carriers of SORT1 variant with diabetes show larger decreases in plasma apoB, TG, and VLDL and LDL particle number. Hepatic sortilin may serve as a quality control step in the apoB secretion pathway.

Latest impact of engineered human liver platforms on drug development

Chase P. Monckton et al.

Summary: Drug-induced liver injury (DILI) is a major cause of drug attrition, and in vitro human liver models are used to assess drug effects and disease modeling more accurately. Bioengineers have developed advanced two-dimensional and three-dimensional human liver platforms using tools from the semiconductor industry and additive manufacturing, enhancing liver cell functions closer to physiologic levels and improving functional lifetime.

Spending on Cardiovascular Disease and Cardiovascular Risk Factors in the United States: 1996 to 2016

Maxwell Birger et al.

Summary: US adult cardiovascular spending increased from $212 billion in 1996 to $320 billion in 2016, mainly driven by increased spending on treatment of hypertension, hyperlipidemia, and atrial fibrillation/flutter. The increase was primarily associated with higher service price and intensity, while changes in disease prevalence contributed to a reduction in spending over the same period.
