Immunoregulation by antigen-presenting cells in human intestinal lamina propria

Takayuki Ogino et al.

Summary: Antigen-presenting cells, such as macrophages and dendritic cells, play a crucial role in the differentiation of T cells and activation of adaptive immune response. Different subsets of these cells in the intestinal lamina propria have been identified and shown to contribute to the maintenance of intestinal homeostasis by interacting with intestinal bacteria. Further research on these antigen-presenting cells in the intestinal tract may help in understanding the pathology of inflammatory bowel disease and developing novel treatment strategies.

Molecular mechanism of the TGF-β/Smad7 signaling pathway in ulcerative colitis

Bingqing Bai et al.

Summary: This study found that anti-TNF-alpha and 5-ASA can improve intestinal epithelial permeability in a mouse model of colitis. The protective effects may be mediated by increased TGF-beta expression and/or reduced Smad7 expression, which inhibit epithelial MLCK activity and decrease mucosal permeability.

The Increase of miR-195-5p Reduces Intestinal Permeability in Ulcerative Colitis, Modulating Tight Junctions' Expression

Viviana Scalavino et al.

Summary: Defects in intestinal epithelial barrier functions are characteristic of inflammatory conditions such as Inflammatory Bowel Disease (IBD). This study found that miR-195-5p can regulate the expression of CLDN2 and other tight junction proteins under inflammatory conditions. It plays an important role in the integrity of the intestinal barrier and epithelial restoration.

miR-195-5p Regulates Tight Junctions Expression via Claudin-2 Downregulation in Ulcerative Colitis

Viviana Scalavino et al.

Summary: This study identified 18 dysregulated miRNAs in intestinal epithelial cells from the ulcerative colitis (UC) mice model. MiR-195-5p, one of the down-regulated miRNAs, was found to be involved in impaired barrier function. The study demonstrated that miR-195-5p can regulate tight junctions by decreasing CLDN2 levels and subsequently affecting CLDN1 expression.

Smad7 Antisense Oligonucleotide-Based Therapy in Crohn's Disease: Is it Time to Re-Evaluate?

Giovanni Monteleone et al.

Summary: This article explores the reasons for the failure of the Phase III clinical trial of mongersen, a drug used to treat Crohn's disease. The study found that different batches of mongersen had chemical differences, with some batches unable to downregulate the expression of Smad7. The research calls for new clinical studies to evaluate the efficacy of chemically homogenous batches of mongersen in patients with inflammatory bowel diseases.

Phosphoinositide-binding activity of Smad2 is essential for its function in TGF-β signaling

Pawanthi Buwaneka et al.

Summary: The study reveals that Smad2 selectively binds to phosphatidylinositol-4,5bisphosphate (PI(4,5)P2) in the plasma membrane, which is crucial for its recruitment and activation at the TGF-il receptor complex. The interaction with PI(4,5)P2 triggers the PM recruitment of Smad2, leading to downstream signaling and transcriptional regulation. The findings suggest a potential role of membrane lipids in regulating Smad proteins and their functions in TGF-il-mediated biological processes.
