Oral pretreatment with β-lactoglobulin derived peptide and CpG co-encapsulated in PLGA nanoparticles prior to sensitizations attenuates cow's milk allergy development in mice

Mengshan Liu et al.

Summary: Cow's milk allergy is a common food allergy among infants. Improved hygiene conditions and loss of microbial diversity are associated with increased risk of allergy development. Oral tolerance induction can be achieved through the administration of PLGA nanoparticles co-encapsulated with beta-lactoglobulin derived peptide and TLR-9 ligand CpG. This treatment prevents allergic reactions and reduces the frequency of Th2 cells in the spleen.

Inhibition of cow's milk allergy development in mice by oral delivery of β-lactoglobulin-derived peptides loaded PLGA nanoparticles is associated with systemic whey-specific immune silencing

Mengshan Liu et al.

Summary: The study demonstrated the efficacy of oral pre-exposure to PLGA nanoparticles loaded with specific peptides in preventing whey-protein induced CMA in mice, showing a partial reduction in symptoms and inhibition of specific immune responses. This suggests potential for further development towards clinical application.

Intestinal Uptake and Tolerance to Food Antigens

Yuhong Xiong et al.

Summary: This article summarizes the mechanisms of food allergy and the lack of individualized treatment options. It also discusses the contribution of gut commensal bacteria to tolerance to food antigens. By elucidating the mechanisms of intestinal uptake and tolerance of food antigens, new clues for the treatment of food allergy are provided.

FRET as the tool for in vivo nanomedicine tracking

Norraseth Kaeokhamloed et al.

Summary: Advanced drug delivery system using nanocarriers is a major application of nanotechnology in pharmacotherapeutics. However, the clinical translation of nanomedicine faces challenges, and this review focuses on utilizing Fo•rster resonance energy transfer (FRET) as a method to track intact nanocarriers in vivo for studying pharmacokinetics.

Implications of Quenching-to-Dequenching Switch in Quantitative Cell Uptake and Biodistribution of Dye-Labeled Nanoparticles

Guangze Yang et al.

Summary: This study compared two types of dye-labeled nanoparticles and found that nanoparticles with dye molecules in a solid state undergo dye quenching, while those in a soluble state do not. To standardize the comparative study, the proposed solutions include using lower dye loading or utilizing medium analysis for quantifying cell uptake of nanoparticles. Valuable insights into selecting valid quantification methods for bio-nano studies are provided.

Summary: The study demonstrates that different sizes of PLGA nanoparticles can affect their uptake by dendritic cells and their trafficking in the skin and lymphoid system. Larger particles are more readily taken up by dendritic cells and are captured and transported more efficiently in vivo.
