A novel definition and treatment of hyperinflammation in COVID-19 based on purinergic signalling

Summary: Hyperinflammation is a crucial factor in severe and critical cases of COVID-19, often characterized by cytokine storm. Treatment with anti-inflammatory drugs has shown limited efficacy and potential serious side effects. The release of cellular ATP by SARS-CoV-2 triggers a pro-inflammatory immune response, leading to further inflammation and immune paralysis. The use of the P2X7R antagonist lidocaine has shown promise in abrogating hyperinflammation and restoring immune function in critically ill COVID-19 patients.

Inhibitors of Human 5-Lipoxygenase Potently Interfere With Prostaglandin Transport

Summary: 5-Lipoxygenase (5-LO) inhibitors show compound-specific pharmacological effects and their efficacy may involve the inhibition of prostaglandin E-2 (PGE(2)) export. Experimental models on the role of 5-LO in inflammation and pain using these inhibitors may be misleading, and their use as pharmacological tools needs to be reevaluated. Additionally, 5-LO inhibitors may serve as new scaffolds for the development of potent prostaglandin export inhibitors.

The newly synthesized compounds showed strong inhibitory activity against tyrosinase, with compound 2n exhibiting the lowest IC50 value. It could be recommended as a potential lead candidate for treating tyrosinase-mediated hyperpigmentation in the future.

In vitro, in vivo and in silico evaluation of the anti-inflammatory potential of Hyssopus officinalis L. subsp. aristatus (Godr.) Nyman (Lamiaceae)

The extracts from hyssop herb showed significant inhibition of COX-2 enzyme in in vitro testing and demonstrated anti-inflammatory effects in in vivo testing. Preliminary in silico testing also showed that the organic acids in hyssop herb had better inhibitory activity against COX-1 and COX-2 enzymes compared to common non-steroidal anti-inflammatory drugs.
