The role of microglia in neuropsychiatric disorders and suicide

Ralf Brisch et al.

Summary: This review examines the potential role of microglial cells in neuroinflammation and neuropsychiatric disorders like schizophrenia, depression, and suicide. It discusses the interactions between microglia, astrocytes, and neurons in pathophysiological processes and explores the effects of drug therapy and electroconvulsive therapy on microglial cells. Finally, it also touches upon the evolutionary role of microglia in human lineage and how this understanding may shape perceptions of neuropsychiatric disorders.

Neuroprotective Effect of Valproic Acid on Salicylate-Induced Tinnitus

Anji Song et al.

Summary: This study investigated the therapeutic effects of valproic acid (VPA) in a salicylate-induced tinnitus model. The results showed that pretreatment with VPA before salicylate treatment can attenuate the increased levels of NMDA receptor, intracellular reactive oxygen species (ROS), and apoptosis-related markers induced by salicylate. These findings provide evidence for the beneficial effects of VPA in the treatment of salicylate-induced temporary hearing loss and tinnitus.

Summary: This study discovers a neuropeptide, KP-10, that can alleviate the damage caused by amyloid-beta and alpha-synuclein in cholinergic neurons. KP-10 mitigates alpha-synuclein-induced toxicity in cholinergic neurons through preferential binding. Moreover, in vitro and in silico experiments support this observation.

Summary: Understanding the transient dynamics of the autonomic nervous system during fear learning is crucial for the treatment of fear-related disorders. In this study, a novel frequency-domain analysis of heart rate was performed to investigate the physiological processes underlying fear learning. The results revealed that fear learning led to a transient heart rate deceleration and increased parasympathetic (vagal) activity, indicating the involvement of cardiac vagal dynamics in fear learning.

Mesenchymal Stem Cells Ameliorate Cuprizone-Induced Demyelination by Targeting Oxidative Stress and Mitochondrial Dysfunction

Elham Shiri et al.

Summary: The transplantation of mesenchymal stem cells (MSCs) is shown to alleviate demyelination in a mouse model of multiple sclerosis (MS) by promoting antioxidant response, reducing oxidative stress, and improving mitochondrial homeostasis. This results in increased myelinated areas and fibers, as well as a decrease in astrogliosis and microgliosis in the corpus callosum. Furthermore, MSC therapy reverses the effects of copper chelator-induced oxidative stress and mitochondrial dysfunction in MS.

Functional Characterization of Spinocerebellar Ataxia Associated Dynorphin A Mutant Peptides

Andreas Lieb et al.

Summary: Mutations in the prodynorphin gene are associated with spinocerebellar ataxia type 23 (SCA23). Pathogenic missense mutations lead to elevated levels of mutant peptide with neurotoxic properties. The study showed that SCA23 DynA mutants decrease the potency of KOR activation, both in terms of G-protein dissociation and beta-arrestin recruitment. Molecular modeling suggests that this loss of function is due to disruption of critical interactions between DynA and the receptor.

Quinolinic acid is associated with cognitive deficits in schizophrenia but not major depressive disorder

Flurin Cathomas et al.

Summary: Tryptophan and its catabolites have been suggested to link immune system activation and neurotransmitter abnormalities with schizophrenia and major depressive disorder. This study found that patients with MDD had lower kynurenine and 3-hydroxy kynurenine levels, while patients with SZ showed a negative correlation between quinolinic acid and cognitive impairment severity. These results suggest dysregulation of the kynurenine pathway in MDD and SZ.

Secondary Progressive Multiple Sclerosis New Insights

Bruce A. C. Cree et al.

Summary: In most cases, multiple sclerosis begins with a relapsing-remitting course followed by secondary progressive MS (SMPS), and there are major differences between RRMS and SPMS in terms of treatment response and diagnosis. However, clear criteria marking the transition from RRMS to SPMS have not been established yet. Early identification of SPMS will require tools that, along with appropriate treatment, may lead to better long-term outcomes.
