The transcriptional regulators of virulence for Pseudomonas aeruginosa: Therapeutic opportunity and preventive potential of its clinical infections

Xiaolong Shao et al.

Summary: In Pseudomonas aeruginosa, transcription factors play a crucial role in the genetic regulation of adaptability and pathogenicity. Several techniques have been used to identify TF binding sites on the P. aeruginosa genome, and an atlas of TF binding patterns has been created. This review summarizes recent literature on TF regulatory networks involved in various processes, including quorum sensing, biofilm formation, pyocyanin synthesis, motility, secretion systems, and oxidative stress responses. Insights from the global regulatory network of transcriptional regulators could inform strategies for preventing clinical infections caused by P. aeruginosa.

How Ethanolic Disinfectants Disintegrate Coronavirus Model Membranes: A Dissipative Particle Dynamics Simulation Study

Tianhang Zhou et al.

Summary: In this study, dissipative particle dynamics models were used to investigate the effects of ethanol on coronavirus model membranes. It was found that 5-10 mol% ethanol significantly weakens the membranes, and the magnitude of the effect depends on the ethanol concentration and membrane composition. The study also revealed that membranes with higher DPPC content are more stable against ethanol and that the presence of ethanol disrupts mixed membranes depending on their composition.

GPU-Accelerated All-Atom Particle-Mesh Ewald Continuous Constant pH Molecular Dynamics in Amber br

Julie A. Harris et al.

Summary: In this study, we developed and validated a GPU-accelerated all-atom continuous constant pH MD method for predicting protonation states in proteins. By using the particle-mesh Ewald (PME) electrostatics in the Amber22 pmemd.cuda engine, we were able to accurately predict pKa values for Asp, Glu, His, and Cys. The method was tested on six benchmark proteins and showed good agreement with experimental results. We also demonstrated that increasing the simulation box size can improve the accuracy of the predictions.

Virtual Screening of Potential Quorum Sensing Inhibitors of P. aeruginosa

Snigdha Bhardwaj et al.

Summary: In this study, potential inhibitors of LasR were identified using molecular docking and the selected compounds were analyzed for their physico-chemical properties and drug likeness. Thiazole derivative was found to have the potential as a novel QSI to combat the virulence of multi-drug resistant P. aeruginosa more effectively.

Pseudomonas aeruginosa: an antibiotic resilient pathogen with environmental origin

Pablo Laborda et al.

Summary: Pseudomonas aeruginosa, known for its low antibiotics' susceptibility, is a relevant opportunistic pathogen causing infections in hospitals and cystic fibrosis patients. It also colonizes environmental ecosystems, with its virulence and antibiotic resistance determinants evolving in nature to play functions beyond infecting human hosts. Although some epidemic clones have been identified, there are no specific lineages linked to infections, highlighting the complexity of P. aeruginosa's population structure.
