Invadosome Formation by Lung Fibroblasts in Idiopathic Pulmonary Fibrosis

Megane Lebel et al.

Summary: Idiopathic pulmonary fibrosis (IPF) is characterized by abnormal fibroblast accumulation in the lung leading to compromised lung function. This study investigated the mechanisms of interstitial invasion and remodeling by lung fibroblasts in IPF and found increased invadosome formation. Additionally, two drugs approved for treating IPF, nintedanib and pirfenidone, were found to suppress invadosome formation in IPF lung fibroblasts.

Epigenetics Approaches toward Precision Medicine for Idiopathic Pulmonary Fibrosis: Focus on DNA Methylation

Wiwin Is Effendi et al.

Summary: Genetic information is not only transmitted by DNA but also through the epigenetics process. Epigenetics plays a crucial role in bridging the gap between genetic background and environmental risk factors in the development of pulmonary fibrosis. Specific epigenetic patterns, such as DNA methylation, histone modifications, long non-coding RNA, and microRNA, have an impact on the pathogenesis of idiopathic pulmonary fibrosis (IPF). Among these, DNA methylation modifications have been extensively studied in IPF. This review summarizes the current understanding of DNA methylation changes in pulmonary fibrosis and highlights the potential of epigenetics-based precision medicine.

Cortical complexity estimation using fractal dimension: A systematic review of the literature on clinical and nonclinical samples

Valentina Meregalli et al.

Summary: Fractal geometry is utilized to characterize the complexity of the brain cortex in various studies focusing on neurodevelopment, aging, and neurobiological aspects of specific psychiatric and neurological disorders. Results suggest that cortical complexity may decrease with aging and various pathological processes, but the high heterogeneity among studies hinders drawing definitive conclusions. Further research integrating FD with other morphological values is necessary to better understand its role in characterizing cortical structure.

Usual interstitial pneumonia (UIP): a clinically significant pathologic diagnosis

Sanjay Mukhopadhyay

Summary: This editorial discusses common issues in the diagnosis of usual interstitial pneumonia (UIP) and emphasizes the distinction between the pathologically defined UIP and the clinically defined idiopathic pulmonary fibrosis (IPF), as well as the determination of etiology and the discrepancy between pathology and high-resolution computed tomography (HRCT) findings.

Usual interstitial pneumonia: a clinically significant pattern, but not the final word

Brandon T. Larsen

Summary: Usual interstitial pneumonia (UIP) is widely used in clinical practice, but its diagnosis lacks a true gold standard. Pathologists need to recognize the limitations of UIP and focus on determining the most likely etiology for targeted therapy. New evidence-based classifications and criteria are required to improve the diagnostic value of biopsies.

Targeting Histone Deacetylases in Idiopathic Pulmonary Fibrosis: A Future Therapeutic Option

Martina Korfei et al.

Summary: Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal lung disease with limited treatment options. Histone deacetylase (HDAC) family of transcriptional corepressors have been identified as crucial mediators of IPF pathogenesis. The imbalance of HDAC activity in IPF lungs promotes and perpetuates fibrosis. HDAC inhibitors show promise as potential therapies for IPF.

TRIB3 Mediates Fibroblast Activation and Fibrosis though Interaction with ATF4 in IPF

Lan Wang et al.

Summary: This study highlights the role of TRIB3 in idiopathic pulmonary fibrosis (IPF), demonstrating its inhibitory effect on fibroblast activation and ECM deposition. TRIB3 negatively regulates ATF4, thereby attenuating pulmonary fibrosis and maintaining normal epithelial-mesenchymal crosstalk.

European Respiratory Society guidelines on transbronchial lung cryobiopsy in the diagnosis of interstitial lung diseases

Daniel A. Korevaar et al.

Summary: This guideline provides evidence-based clinical practice recommendations for the role of transbronchial lung cryobiopsy (TBLC) in obtaining tissue-based diagnosis in patients with undiagnosed interstitial lung diseases (ILD). TBLC provides important diagnostic information with lower diagnostic yield compared to surgical lung biopsy (SLB), at reduced serious adverse events and length of hospitalisation.

Digital quantification of p16-positive foci in fibrotic interstitial lung disease is associated with a phenotype of idiopathic pulmonary fibrosis with reduced survival

Jonathan Keow et al.

Summary: This study found that p16-positive foci can be used to diagnose idiopathic pulmonary fibrosis and are not related to radiographic patterns. High p16 expression is associated with shorter survival time, while antifibrotic therapy has a significant protective effect. Digital spatial profiling showed that fibroblastic foci have distinct gene expression compared to normal and fibrotic areas.

Kati Makela et al.

Summary: This study aimed to use artificial intelligence to quantitatively analyze fibroblast foci, interstitial mononuclear inflammation, and intra-alveolar macrophages in lung tissue samples of patients with idiopathic pulmonary fibrosis (IPF). The deep convolutional neural network detected histological features that are difficult to quantitate manually and identified novel prognostic histological markers in IPF, providing new information for prognostic estimation of the disease.
