Activation of NOTCH signaling via DLL1 is mediated by APE1-redox-dependent NF-κB activation in oesophageal adenocarcinoma

Lei Chen et al.

Summary: This study investigates the role of acidic bile salts in activating NOTCH signaling in oesophageal adenocarcinoma (EAC). The researchers found that there is a crosstalk between APE1 redox function, reflux-induced inflammation, and DLL1 upregulation, which promotes cancer cell stem-like properties in EAC. The overexpression of APE1 and DLL1 is associated with poor overall survival in EAC patients.

Identification of visible lesions during surveillance endoscopy for Barrett's esophagus: a video-based survey study

Claire A. Beveridge et al.

Summary: This study aimed to assess the rate of visible lesion (VL) detection by academic and community endoscopists using high-definition white-light endoscopy (HD-WLE) and narrow-band imaging (NBI). Participants identified VLs using HD-WLE and NBI, and the results showed that 28% and 31% of VLs were not identified using HD-WLE and NBI respectively. The study also found that the rate of VL detection was associated with the endoscopist's experience and the number of surveillance endoscopies performed each month.

Shyam Vedantam et al.

Summary: In this study, 9 studies were identified, involving 2356 patients with Lynch syndrome who underwent 7838 upper endoscopy screenings. The results showed that upper GI cancer screening using endoscopy can detect Lynch syndrome-associated upper GI malignancies, high-risk lesions, and other clinically actionable findings, supporting its use as part of cancer risk management in Lynch syndrome.

Nicholas J. Shaheen et al.

Summary: This study evaluated the risk of progression in different types of Barrett's esophagus (BE) reported by WATS3D, including crypt dysplasia (CD) and low-grade dysplasia (LGD). The results showed that non-dysplastic BE diagnosed by WATS3D had a very low risk of progression, while CD had a significantly higher risk compared to non-dysplastic BE but lower than LGD.

Summary: The aim of this study was to evaluate the value of p53, Ki67, and toll-like receptor 5 (TLR5) in predicting the malignant progression of Barrett's esophagus. It was found that aberrant p53 expression is associated with a high risk of malignant progression in Barrett's low-grade dysplasia. Routine staining of p53 is recommended in expert confirmed low-grade dysplasia cases.

Endoscopic Diagnosis and Management of Barrett's Esophagus with Low-Grade Dysplasia

Francesco Maione et al.

Summary: Barrett's Esophagus is a common condition associated with chronic gastroesophageal reflux disease and has a higher risk of developing esophageal adenocarcinoma. Low and high-grade dysplasia precede neoplastic transformation in this condition. The evaluation of low-grade dysplastic esophageal mucosa is still controversial, but endoscopic surveillance and minimally invasive endoscopic treatments such as endoscopic mucosal resection and radiofrequency ablation are available options. Cryotherapy ablation and argon plasma coagulation are novel endoscopic treatments with high eradication rates and fewer complications and post-procedural pain.

The Ephrin B2 Receptor Tyrosine Kinase Is a Regulator of Proto-oncogene MYC and Molecular Programs Central to Barrett's Neoplasia

Srividya Venkitachalam et al.

Summary: The study suggests that the EphB2 signaling pathway is frequently hyperactivated in the early development of Barrett's esophagus-associated esophageal adenocarcinoma (BE-EAC) and acts as an upstream regulator of the MYC gene. Inhibiting EphB2 can significantly impede the proliferation of BE and EAC cells. Targeting EphB2/MYC could be a promising strategy for treating this common refractory and aggressive cancer.

Development of a deep learning model for the histologic diagnosis of dysplasia in Barrett's esophagus

Shahriar Faghani et al.

Summary: A deep learning model was developed to accurately predict the dysplasia grade of Barrett's esophagus (BE) on whole-slide images. The model, using an ensemble approach, demonstrated high sensitivity and specificity for low-grade dysplasia (LGD), non-dysplastic BE (NDBE), and high-grade dysplasia (HGD) diagnoses. The findings of this study are important for improving the histologic diagnosis of BE dysplasia and the appropriate use of endoscopic therapy.

An anti-CTLA-4 heavy chain-only antibody with enhanced Treg depletion shows excellent preclinical efficacy and safety profile

Xin Gan et al.

Summary: The value of anti-CTLA-4 antibodies in cancer therapy is well established. However, current anti-CTLA-4 therapeutic antibodies have limitations that hinder their broad application. This study successfully developed HCAb 4003-2, a fully human heavy chain-only antibody, which showed superior antitumor activity compared to ipilimumab and other anti-CTLA-4 antibodies in in vitro and in vivo experiments. HCAb 4003-2 also demonstrated a shorter serum half-life and lower systemic drug exposure. These findings make HCAb 4003-2 a promising candidate for the development of next generation anti-CTLA-4 therapy.

International trends in esophageal cancer incidence rates by histological subtype (1990-2012) and prediction of the rates to 2030

Yushi Lin et al.

Summary: The incidence rates of esophageal squamous cell carcinoma (ESCC) have been decreasing worldwide, while the rates of esophageal adenocarcinoma (EAC) have been increasing. Most countries are expected to continue experiencing a decreasing or stable trend in esophageal cancer incidence, except Denmark, the Netherlands, and the UK where the rates of EAC are predicted to increase. The changing patterns of these two subtypes of esophageal cancer may require innovative interventions to address issues such as obesity, GERD, and Barrett's esophagus.

Sex Differences in Neoplastic Progression in Barrett's Esophagus: A Multicenter Prospective Cohort Study

Carlijn A. M. Roumans et al.

Summary: Barrett's esophagus (BE) is the precursor lesion of esophageal adenocarcinoma (EAC). The cost-effectiveness of current surveillance strategies for BE is debatable. This multicenter prospective cohort study aimed to evaluate sex differences in 868 BE patients. The results showed that males had higher risk and shorter time to neoplastic progression, while females were more likely to be diagnosed with (advanced) EAC. These findings can guide the development of sex-specific guidelines and improve neoplastic risk stratification in BE surveillance.

Immunotherapy in triple-negative breast cancer: Insights into tumor immune landscape and therapeutic opportunities

Rita Ribeiro et al.

Summary: Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer that is more common in young pre-menopausal women. This review discusses the implications of TNBC's characteristics, such as its lack of hormone receptors and high tumor mutational burden, for diagnosis, treatment, and prognosis. It highlights the potential of immunotherapy as an alternative treatment for TNBC, including the use of immune gene signatures and tumor-infiltrating lymphocytes as biomarkers. The review also explores different formats of immunotherapy currently under clinical research, such as immune checkpoint inhibitors and antibody-drug conjugates.
