Managing expectations, rights, and duties in large-scale genomics initiatives: a European comparison

Ruth Horn et al.

Summary: This article reports the findings of an international workshop organized by UK-France Genomics and Ethics Network (UK-FR GENE) in 2021. The workshop focuses on the challenges posed by the collection, storage, and sharing of genomic data in countries implementing large-scale national genomic initiatives. Comparative analysis includes initiatives in England, France, and Germany, and highlights the need for careful management and transparent governance structures for biomedical data. The article concludes that while the potential for improving patient healthcare and contributing to research is undeniable, patient benefits must outweigh commercial interests and cross-border data transfer must address regulatory inconsistencies.

Indications and Limitations of Sirolimus in the Treatment of Vascular Anomalies-Insights From a Retrospective Case Series

Anna Karastaneva et al.

Summary: This study retrospectively analyzed the use of sirolimus in treating vascular anomalies in 16 children between 2014 and 2020. It found that sirolimus can be effective in reducing volume and improving vascular anomalies, but its use as a monotherapy may have limitations. Combination regimens and multimodal treatment strategies should be explored. Sirolimus's side effects include mucositis and laboratory abnormalities.

Trametinib as a promising therapeutic option in alleviating vascular defects in an endothelial KRAS-induced mouse model

H-L Nguyen et al.

Summary: The study reports the association between KRAS mutations and arteriovenous malformations. By activating KRAS in vascular endothelial cells using a mouse model, the researchers successfully generated an arteriovenous malformation mouse model. They also found that trametinib could effectively slow down the progression of the lesions and has the potential to be a therapeutic option for KRAS-induced vascular malformations.

Case report: five-year experience of AKT inhibition with miransertib (MK-7075) in an individual with Proteus syndrome

Christopher A. Ours et al.

Summary: Proteus syndrome, a rare overgrowth disorder, can cause functional impairment and disfigurement. Treatment options are limited, but AKT inhibitors like miransertib show promise in mitigating the morbidity of this syndrome, as evidenced in a five-year follow-up study of a single patient. Further research is needed to confirm the safety and effectiveness of this targeted pharmacologic approach.
