Seed amplification assay for the detection of pathologic alpha-synuclein aggregates in cerebrospinal fluid

Luis Concha-Marambio et al.

Summary: Misfolded alpha-synuclein aggregates can be amplified in vitro using alpha Syn seed amplification assays, which have shown promise as a biochemical test for synucleinopathies. This assay can detect alpha Syn aggregates in cerebrospinal fluid samples from patients with Parkinson's disease, dementia with Lewy bodies, or multiple-system atrophy, and distinguish between samples from patients with Parkinson's disease and those with multiple-system atrophy.

Alpha-synuclein seeding shows a wide heterogeneity in multiple system atrophy

Ivan Martinez-Valbuena et al.

Summary: This study revealed significant differences in the seeding activity of alpha-synuclein in the brains of patients with Multiple System Atrophy (MSA), as well as regional variations within individual brains. These findings provide important experimental groundwork for future subclassification and rapid diagnostic assays for MSA.

Effect of olfactory bulb pathology on olfactory function in normal aging

Cecilia Tremblay et al.

Summary: The decline of olfactory function is frequently observed in aging. This study found that tau pathology is commonly observed in the olfactory bulb of normally aging individuals, but it does not independently contribute to age-related olfactory decline. Further involvement of the brain seems to be necessary for age-related olfactory impairment.

Association between probable REM sleep behavior disorder and increased dermal alpha-synuclein deposition in Parkinson's disease

K. Doppler et al.

Summary: In patients with Parkinson's disease, the presence of REM sleep behavior disorder is associated with higher frequency and abundance of dermal alpha-synuclein deposition, resembling those with isolated REM sleep behavior disorder. This suggests a potential link between REM sleep behavior disorder and a specific subtype of Parkinson's disease characterized by peripheral nerve involvement.

Alpha-Synuclein Strain Variability in Body-First and Brain-First Synucleinopathies

Mie Kristine Just et al.

Summary: Pathogenic alpha-synuclein aggregates are a defining feature of neurodegenerative synucleinopathies. Early differentiation between these diseases is challenging due to clinical heterogeneity. However, recent advancements in seed amplification assays and conformation-specific ligand assays show promise in differentiating synucleinopathies.

A postmortem study suggests a revision of the dual-hit hypothesis of Parkinson's disease

Per Borghammer et al.

Summary: The dual-hit hypothesis of Parkinson's disease suggests that a neurotropic pathogen leads to formation of alpha-synuclein pathology that enters the brain from the olfactory bulb and the dorsal motor nucleus of the vagus. However, a re-analysis of postmortem datasets found that early body-first Lewy body disease (LBD) cases rarely had olfactory bulb pathology, while early brain-first LBD cases almost always showed olfactory bulb pathology. This supports the idea that the pathological process starts in either the olfactory bulb or the enteric nervous system, but not both simultaneously.

Olfactory swab sampling optimization for α-synuclein aggregate detection in patients with Parkinson's disease

Summary: This study found that nasal swab testing performed at the agger nasi significantly increased the sensitivity of real-time quaking-induced conversion (RT-QuIC) detection of pathological alpha-synuclein in patients with Parkinson's disease (PD). The study also observed the presence of alpha-synuclein and phospho-alpha-synuclein deposits in nasal swabs from PD patients.

Opportunities and challenges of alpha-synuclein as a potential biomarker for Parkinson's disease and other synucleinopathies

Pedro Magalhaes et al.

Summary: This review provides an overview of the distribution of alpha-synuclein (aSyn) species in the brain, peripheral tissues, and biological fluids. It analyses previous studies on measuring total aSyn and other modified and aggregated forms of aSyn in different biological fluids. It highlights the conceptual and technical gaps and challenges in developing and validating reliable aSyn biomarkers and proposes recommendations to address these challenges.

Skin α-Synuclein Aggregation Seeding Activity as a Novel Biomarker for Parkinson Disease

Zerui Wang et al.

Summary: The study demonstrates that skin alpha Syn(P) seeding activity can potentially serve as a novel biomarker for antemortem diagnosis of Parkinson disease (PD) and other synucleinopathies. The analysis of skin samples from autopsies and biopsies shows a significant increase in alpha Syn(P) seeding activity in individuals with PD and synucleinopathies compared to controls with tauopathies and non-neurodegenerative diseases.

Neuropathological evidence of body-first vs. brain-first Lewy body disease

Per Borghammer et al.

Summary: The aggregation of alpha-synuclein into inclusion bodies, known as Lewy pathology, is a key feature of Parkinson's disease and Dementia with Lewy bodies. Post mortem studies have shown that Lewy pathology follows two main distribution patterns in the brain, with some patients having pathogenic alpha-synuclein originating in the enteric nervous system and others originating inside the central nervous system.

Prodromal Parkinson disease subtypes - key to understanding heterogeneity

Daniela Berg et al.

Summary: Parkinson's disease begins its pathological processes and neurodegeneration long before clinical diagnosis, allowing for identification of individuals likely to develop the disease through risk and prodromal markers. Heterogeneity in clinical and prodromal PD suggests that subtypes exist, differentiated by clinical manifestations, pathomechanisms, and progression patterns. Understanding this variability is crucial for early and accurate diagnosis, as well as for targeting neuroprotective interventions effectively.
