Oxidative Stress and Oocyte Cryopreservation: Recent Advances in Mitigation Strategies Involving Antioxidants

Beijia Cao et al.

Summary: Oocyte cryopreservation induces oxidative stress and reduces the developmental potential of oocytes. While oocytes have intrinsic antioxidant systems, cryopreservation still causes oxidative damage. Exogenous antioxidants, such as melatonin and resveratrol, can reduce oxidative damage. This review discusses the analysis of oxidative stress induced by oocyte cryopreservation, the impact of antioxidants against oxidative damage, and their underlying mechanisms, providing a reference for improving the efficiency of oocyte cryopreservation.

Calcium (Ca2+) expression and intensity in cumulus-oocyte complex (COCs ) in Kacang goat after vitrification

W. Widjiati et al.

Summary: The process of cryopreservation affects the expression and intensity of calcium (Ca2+) in cumulus-oocyte complexes (COCs). Cryopreservation disrupts the balance of intracellular calcium and increases the presence of Reactive Oxygen Species (ROS), leading to changes in Ca2+ levels. The intensity of Ca2+ is concentrated in the nucleus of COCs after vitrification, while in fresh COCs it is concentrated in the cytoplasm.

Role of astaxanthin as an efficient antioxidant on the in vitro maturation and vitrification of porcine oocytes

De-Cai Xiang et al.

Summary: The study demonstrated that Astaxanthin (Ax) significantly improved the survival rate of vitrified oocytes, promoted blastocyst yield, reduced reactive oxygen species generation, and increased glutathione level. Ax also decreased cathepsin B activity, increased lysosome fluorescence intensity, enhanced mitochondrial activity, and restored expression of specific genes in vitrified oocytes, ultimately improving developmental competence.

Mitochondrial Dysfunction and Oxidative Stress Caused by Cryopreservation in Reproductive Cells

Roberto Gualtieri et al.

Summary: Mitochondria play a crucial role in cell metabolism and ATP synthesis, while also being responsible for generating ROS and maintaining calcium homeostasis. Dysfunction in mitochondria can lead to oxidative stress, affecting cell function. Assisted reproductive technologies and cryopreservation may compromise mitochondrial function, impacting reproductive outcomes by affecting sperm and oocyte quality and fertilization dynamics.

Oxidative stress in oocyte aging and female reproduction

Ling Wang et al.

Summary: Oxidative stress plays a significant role in oocyte aging, leading to a gradual decrease in oocyte quantity and quality, as well as various reproductive pathologies. Strategies to delay oocyte aging include targeting oxidative stress through autophagy antioxidant pathways and antioxidants, along with creating a hypoxic culture environment for oocytes in vitro.

Melatonin improves the first cleavage of parthenogenetic embryos from vitrified-warmed mouse oocytes potentially by promoting cell cycle progression

Bo Pan et al.

Summary: Our study revealed that the cryopreservation of oocytes can lead to spindle abnormalities, mitochondrial dysfunction, increased DNA damage, and ultimately affect embryo development. However, supplementation of melatonin (MT) can alleviate these negative impacts and promote cell cycle progression, enhancing the first cleavage and subsequent development of parthenogenetic zygotes developed from vitrified-warmed oocytes.
