Chemogenetic activation of astrocytes promotes remyelination and restores cognitive deficits in visceral hypersensitive rats

Summary: In this study, a rat model of chronic visceral hypersensitivity (VH) was used to investigate the role of myelination in impaired decision-making and network desynchronization. The findings showed a decrease in myelin basic protein and reduced number of mature oligodendrocytes in the anterior cingulate cortex (ACC) of VH rats. Astrocytic Gq modulation was found to enhance oligodendrocyte proliferation and myelination in the ACC, improving decision-making and synchronization. These insights have potential implications for myelin-related diseases such as chronic pain-associated cognitive disorders.

However, the mechanisms underlying these effects are not fully understood. Novel research tools have helped redefine the locus coeruleus system and have shown its functional heterogeneity. This review summarizes the available data on the efferent activity of the locus coeruleus-noradrenergic system in relation to pain and its comorbidity with anxiodepressive disorders.
