Enhancing a SARS-CoV-2 nucleocapsid antigen test sensitivity with cost efficient strategy through a cotton intermembrane insertion

Diego Rinaldi Pavesi Nicollete et al.

Summary: The aim of this study was to create a lateral flow test that could detect SARS-CoV-2 nucleocapsid protein in low concentrations comparable to existing tests. The research successfully increased the sensitivity by adjusting the test line antibody concentration and adding an intermembrane, resulting in a new rapid test prototype called lateral flow intermembrane immunoassay test (LFIIT).

Quantitative SARS-CoV-2 Viral-Load Curves in Paired Saliva Samples and Nasal Swabs Inform Appropriate Respiratory Sampling Site and Analytical Test Sensitivity Required for Earliest Viral Detection

Emily S. Savela et al.

Summary: Early detection is crucial in controlling the spread of SARS-CoV-2. This study found that high-analytical-sensitivity saliva testing could detect infection several days earlier than low-analytical-sensitivity nasal-swab testing. Although nasal swabs had higher peak viral loads, they were undetectable or at lower loads during the initial days of infection. These findings emphasize the importance of acquiring early viral-load profiles to guide testing strategies and response to emerging variants.

A Rapid RT-LAMP Assay for SARS-CoV-2 with Colorimetric Detection Assisted by a Mobile Application

Maria Aurora Londono-Avendano et al.

Summary: Loop-mediated amplification (LAMP) has been promoted for SARS-CoV-2 screening, but antigen tests are preferred in low-income countries and remote zones. In this study, an in-house LAMP assay with colorimetric detection achieved by the naked eye and a mobile application was used. The accuracy of detection improved when using the application, and saliva samples showed potential for further improvement.

Giulia De Angelis et al.

Summary: The study used antigen detection tests on samples from symptomatic and asymptomatic COVID-19 patients to determine the activity level of SARS-CoV-2 infection. The results showed that different tests had varying levels of sensitivity and specificity in detecting active replication of the virus.

Inhibition of Non-specific Amplification in Loop-Mediated Isothermal Amplification via Tetramethylammonium Chloride

MinJu Jang et al.

Summary: Loop-mediated isothermal amplification (LAMP) is a molecular and point-of-care diagnostic technique used for pathogen detection, but non-specific amplification often occurs. In this study, we discovered that tetramethylammonium chloride (TMAC) can effectively suppress non-specific amplification in LAMP by using additives.

Development of multiplexed reverse-transcription loop-mediated isothermal amplification for detection of SARS-CoV-2 and influenza viral RNA

Yinhua Zhang et al.

Summary: This study enhances loop-mediated isothermal amplification through multiplexed real-time and end point detection of SARS-CoV-2 combined with influenza and control targets. By enabling multiple target detection, loop-mediated isothermal amplification can be even more widely used for diagnostics in settings where multiple viral targets are potential infectious agents and where higher-throughput testing is advantageous.
