Theoretical study on the free radical scavenging potency and mechanism of natural coumestans: Roles of substituent, noncovalent interaction and solvent

Summary: The free radical scavenging potency and mechanisms of seven natural coumestans were evaluated using density functional theory. The results showed that these compounds prefer formal hydrogen atom transfer mechanism in non-polar environment and gas phase, while sequential proton loss electron transfer mechanism is favored in polar media. Sequential double proton loss double electron transfer mechanism represents the preferred pathway in aqueous solution at physiological pH. The essential role of ortho-dihydroxyl group, noncovalent interaction, and solvents on radical-trapping potency was highlighted.

Computational analysis of peroxyl radical scavenging capacity of coumestrol: insights into kinetics and reaction mechanisms

Summary: This study evaluated the scavenging activity of Coumestrol on peroxyl radicals and found that it is influenced by the environment and the nature of the radicals. In lipid media, Coumestrol exhibited moderate scavenging activity through the formal hydrogen transfer mechanism. In polar environments, its activity became more important and tended to use the single electron transfer mechanism.

Tautomerism and antioxidant power of sulfur-benzo[h]quinoline: DFT and molecular docking studies

Summary: In this study, a quantum chemical investigation was conducted to explore the antiradical activity of sulfur-benzo[h]quinolone in physiological environments. The results showed that sulfur-benzo[h]quinolone exhibits solvent-dependent tautomerism and possesses higher antioxidant activity compared to reference antioxidants. Additionally, it was found to be a potential inhibitor of several enzymes.

Summary: The study investigated the in silico ability of a new compound called Wal to act as a hydroperoxyl radical scavenger and pro-oxidant enzyme inhibitor. The results showed that the main antioxidant mechanism of Wal is the formal hydrogen transfer from the OH group. In addition, Wal has greater scavenging activity against the HOO center dot radical compared to the reference antioxidant Trolox, and it can inhibit the activity of various enzyme families.

The radical scavenger capacity and mechanism of prenylated coumestan-type compounds: a DFT analysis

Summary: This study examined the antiradical capacity and mechanisms of two representative coumestan-type compounds using quantum chemistry calculations and computational kinetics methods. It was found that the 18CH groups of the prenyl substituent are the most suitable sites for radical attacks from a thermodynamic perspective. However, kinetic study revealed that the reaction at these CH groups is slow and does not significantly contribute to the overall reactivity of these compounds.

Antioxidant activity of Trolox derivatives toward methylperoxyl radicals: thermodynamic and kinetic theoretical study

Summary: The study explored the thermodynamic and kinetic aspects of Trolox derivatives reactions with methylperoxyl radical CH3OO· using density functional theory. It found that HAT is the preferred mechanism in lipid media while SPLHAT is the most favored mechanism in water. The calculated rate constants showed that the antioxidant activities of the Trolox derivatives are significantly faster than that of the reference system Trolox.
