Preferential Solvation Study of the Synthesized Aldose Reductase Inhibitor (SE415) in the {PEG 400 (1) + Water (2)} Cosolvent Mixture and GastroPlus-Based Prediction

Summary: In this study, a binary cosolvent system of PEG 400 + water was used to enhance the solubility of (Z)-N-Benzyl-2-{2,4-dioxo-5-(4- prop-2-yl-1-yloxyl)benzylidene)thiazolin-3-yl)}acetamide (SE415). The preferential solvation of SE415 by PEG 400 was investigated using Kirkwood-Buff integrals and GastroPlus-based predictions. The results showed that PEG 400 had the highest solubilizing effect on SE415, and the in vitro and in vivo performance of SE415 were predicted to be improved using this solvent system.

Summary: The newly synthesized compound SE11 has been found to inhibit aldose reductase and effectively control diabetes mellitus and related complications. However, SE11 exhibits poor water solubility. This study explores the use of NMP as a co-solvent and investigates the preferential solvation of SE11 by mixed NMP-water solutions, as well as its in vitro dissolution and simulation.

The highest preferential solvation parameter by PEG was observed in the mixture with x(1) = 0.15, being 1.55 x 10(-2).

Rosuvastatin cocrystals: an attempt to modulate physicochemical parameters

Summary: This study aimed to improve the physicochemical properties of rosuvastatin calcium by preparing cocrystals, resulting in enhanced solubility and dissolution rate through establishing intermolecular interactions. The novel cocrystals of rosuvastatin with L-asparagine and L-glutamine showed improved characteristics and demonstrated the benefits of co-crystallization for drug properties enhancement.
