Perinatal brain damage - what the obstetrician needs to know

Summary: Perinatal brain damage is a major cause of perinatal death and postnatal disability worldwide, but it includes various factors that result in insult to the developing brain without differentiation. Hypoxic-ischemic encephalopathy, intraventricular hemorrhage, periventricular leukomalacia, and perinatal stroke are commonly identified as the main causes of perinatal brain damage, with differences in injury type, timing, neuropathological and imaging findings, and clinical presentation. Each type of injury appears to have a specific vulnerability window along the timeline of neurodevelopment in utero, although there is clinical overlap. Previously, peripartum acute hypoxia was considered the major cause, but intrauterine inflammation, prematurity, chronic hypoxia/growth retardation, and genetic abnormalities are also important contributors.

Mitochondrial fusion regulates proliferation and differentiation in the type II neuroblast lineage in Drosophila

Summary: Mitochondrial morphology and function play crucial roles in stem cell differentiation and organism development. Depleting mitochondrial fusion proteins in Drosophila neuroblasts led to fragmentation and loss of activity, affecting differentiation. Mutants in mitochondrial fission protein did not affect differentiation. Disruption of Notch signaling pathway led to fragmentation, while its upregulation resulted in clustering of mitochondria. Crosstalk between proliferation, Notch signaling, mitochondrial activity, and fusion are essential for differentiation in neuroblast lineages.

The turnover of mitochondria, including mitophagy and biogenesis, is critical for brain development and functional recovery. Excessive mitophagy in the early phase may be pathological, leading to secondary energy depletion, while secondary mitophagy may contribute to post-HI regeneration and repair.
