Engineering of a T7 Bacteriophage Endolysin Variant with Enhanced Amidase Activity

Jaya Lakshmi Tyagi et al.

Summary: The therapeutic use of bacteriophage-encoded endolysins as enzybiotics has been significantly increased due to the emergence of antibiotic resistant bacteria. This study employed a structure-guided mutagenesis approach to engineer a T7 bacteriophage endolysin with enhanced amidase activity and lysis potency. The results provided deeper insights into the structure-function relationships of endolysin proteins and aided in the rational design of engineered endolysins with enhanced therapeutic properties.

The Major Autolysin Atl Regulates the Virulence of Staphylococcus aureus by Controlling the Sorting of LukAB

Xuhui Zheng et al.

Summary: Infections caused by Staphylococcus aureus, a Gram-positive bacterium, are a significant global health threat. The production of leukocidins, which are pore-forming toxins, plays a crucial role in the pathogenesis of S. aureus. The transcriptional regulation of these toxins is well-studied, but the posttranslational regulation is not well explored. This study reveals that a major cell wall hydrolase, autolysin (Atl), controls the sorting of one of the leukocidins, LukAB, from the cell envelope to the extracellular milieu. This sorting effect is independent of transcriptional regulation. Importantly, Atl's activity influences S. aureus cytotoxicity toward human neutrophils. The study also shows that peptidoglycan cleavage plays a role in the sorting of S. aureus toxins during secretion.

Summary: By studying the different genotypes of Staphylococcus aureus in humans and cows, it was determined that some predominant human genotypes have adapted to or caused occasional infections in cows. Comparative genomics revealed differences in the number of pseudogenes between RST4-1 and RST2-1 strains, with most pseudogenes being strain-specific.

Development of Advanced Chimeric Endolysin to Control Multidrug-Resistant Staphylococcus aureus through Domain Shuffling

Chanyoung Lee et al.

Summary: By swapping different domains of natural Staphylococcus endolysins, a novel chimeric endolysin ClyC was constructed, showing enhanced lytic activity against S. aureus, strong antibacterial effects in milk and blood, and effective eradication of biofilms of multidrug-resistant bacteria. The chimeric endolysin also demonstrated protection against MRSA in a mouse infection model, suggesting its potential as an antibacterial agent against multidrug-resistant S. aureus.
